树灵芝中三萜类成分及其保肝作用研究

Triterpenoids of Ganoderma sessile and Their Hepatoprotective Activities

摘要: 研究树灵芝Ganoderma sessile中三萜类成分及其保肝作用。树灵芝乙醇提取物经凝胶色谱、硅胶色谱、硅胶薄层色谱、C-18反相硅胶色谱、高效液相色谱和反相制备色谱等色谱技术分离纯化,通过各种波谱学方法鉴定化合物的结构。从树灵芝中分离得到16个三萜类化合物,分别为灵芝酸LM-2(1)、灵芝孢子酸A(2)、灵芝酸J(3)、灵芝酸C(4)、灵芝酸A(5)、灵芝酸B(6)、灵芝酸M(7)、灵芝酸C6(8)、灵芝酸AM1(9)、灵芝酸G(10)、灵芝酸I(11)、灵芝赤芝酸A(12)、灵芝酸ε(13)、灵芝酸H(14)、灵芝赤芝酸E(15)和灵芝酸D(16)。化合物1~16均为首次从该种植物中分离得到。其中化合物2、5、6、11、15和16对人肝HL-7702细胞的成活率分别为60%、45%、56%、44%、47%和47%,均高于双环醇42%的成活率,具有肝保护的作用。

Abstract: Sixteen lanostane triterpenoids were isolated from the ethanol extract of Ganodermasessile by means of preparative HPLC,column chromatography over sephadex LH-20,silica gel,ODS and were identified as ganoderic acid LM-2 (1),ganosporeric acid A (2),ganoderic acid J (3),ganoderic acid C (4),ganoderic acid A (5),ganoderic acid B (6),ganoderic acid M (7),ganoderic acid C-6 (8),ganoderic acid AM-1(9),ganoderic acid G(10),ganoderic acid I(11),ganolucidic acid A (12),ganoderic acid ε(13),ganoderic acid H(14),ganolucidic acid E (15),ganoderic acid D (16).Compounds 1-16 were isolated from this Species for the first time.Compounds 2,5,6,11,15 and 16 exhibited hepatoprotective activities.The survival rate were 60%,45%,56%,44%,47% and 47% in HL-7702 cells respectively,all higher than 42% of bicyclol.
