藏药细果角茴香总生物碱体内抗炎镇痛活性及其质量控制

Study on Anti-Inflammatory,Analgesic Activity and Methods of Quality Control of Total Alkaloids in Hypecoum leptocarpum Hook.f.et Thoms

摘要: 本文选用MCI 微孔树脂对细果角茴香醇提部位进行纯化富集,得到细果角茴香总生物碱,分别考察其总生物碱对小鼠福尔马林致痛和角叉菜胶致小鼠足肿胀的影响;同时采用硅胶GF254薄层板,以氯仿甲醇(6:15)为展开剂,碘化铋钾试液显色,进行细果角茴香总生物碱的TLC鉴别;以原阿片碱为指标性成分,采用RPHPLC法测定其含量。结果表明:细果角茴香总生物碱具有明显的抗炎镇痛活性,原阿片碱在0.96 ~ 4.8 μg范围内与峰面积呈良好的线性关系,回归方程为:A = 6.37 × 106 X + 6.24×105,r=0.9999。以原阿片碱为对照品,HPLC法测定细果角茴香总生物碱的含量,TLC法进行有效成分鉴别,方法简便、易行、结果可靠,可作为细果角茴香抗炎镇痛有效部位的质量控制方法。

Abstract: Total alkaloids in Hypecoum leptocarpum Hook.f.et Thoms.were obtained from the ethanol extract using MCI gel.The antiinflammatory and analgesic effects of total alkaloids were studied using carrageenan induced edema and formalin test.The TLC identification of total alkaloids was carried out using GF254 silica gel plate with developing solvent of chloroformmethanol (6:1.5).The content of total alkaloids were determined by RPHPLC taking protopine as the index component.The results showed that total alkaloids in H.leptocarpum have edema inhibition and analgesic effect.The content of protopine in the total alkaloids was up to 13.3%.The calibration curve was in the range of 0.96-4.8 μg and the equation of regression was A = 6.37 × 106 X + 6.24×105,r=0.9999.The HPLC TLC method were accurate and simple.They can be used for the quality control of this active fraction in H.leptocarpum.
