超高效液相色谱法测定不同种质资源五味子藤茎中5种黄酮类成分的含量

Simultaneous Determination of Five Flavonoids Ingredients in the Stems of Schisandra chinensis from Different Germplasm Resources by UPLC

摘要: 建立超高效液相色谱法同时检测五味子藤茎中芦丁、金丝桃苷、槲皮素-3-O-葡萄糖、槲皮素-3-O-木糖和槲皮素的含量,并比较不同种质资源五味子藤茎中5种黄酮类成分的含量差异。采用ACQUITY UPLC BEH C18(2.1 mm×5.0 mm,1.7 μm)色谱柱,0.1‰磷酸水溶液-乙腈进行梯度洗脱,检测波长为360 nm流速为0.35 mL/min,柱温为30 ℃。在该色谱条件下,五味子藤茎中的5种黄酮类成分能得到较好的分离,方法的加标回收率在96.83%~102.2%,RSD<3%。19种不同种质资源间五味子藤茎的黄酮类成分含量存在较大差异,其中94-9-2、130-9-3和99-8-7显著优于其他品系,94-9-2品系最佳。该方法操作简便,可为五味子藤茎的质量控制提供参考,同时为五味子的种质资源收集和品种选育提供理论参考。

Abstract: An UPLC method was established for simultaneous determination of five flavonoids in the stems of Schisandra chinensis,including rutin,hyperoside,quercetin-3-O-glucoside,quercetin-3-O-xyloside and quercetin.This method was then used to determine the contents of flavonoids in the stems of S.chinensis from different germplasm resources.The five compounds were analyzed simultaneously with an ACQUITY UPLC BEH C18(2.1 mm×5.0 mm,1.7 μm)column by gradient elution using 0.1‰ (v/v) phosphoric acid-acetonitrile as the mobile phase at the flow rate of 0.35 mL/min;The detection wavelength was set at 360 nm,and the column temperture was maintained at 30 ℃.The five flavonoids ingredients of S.chinensis can be separated in the chromatographic conditions.The recoveries of five components were 96.83%~102.15%,with RSD less than 3%.The contents of flavonoids in the stems of S.chinensis  from 19 different germplasm resources had significant differences;94-9-2、130-9-3 and 99-8-7 were significantly better than other strains,94-9-2 was the best.This method was easy to operate and could provide a reference for the quality control of the stem of S.chinensis,and provided theoretical reference for the collection and breeding of S.chinensis germplasm resources.
