Microwave Assisted Extraction and Immunomodulatory Effects of Pomegranate Peel Polysaccharide

摘要: 优化石榴皮多糖的微波辅助提取工艺并初步探究石榴皮多糖的免疫调节作用。在单因素试验的基础上,以液料比、微波提取时间、微波功率为自变量,石榴皮多糖得率为响应值,利用响应曲面分析方法,确定石榴皮多糖提取工艺的最佳条件为:液料比44∶1 mL/g,微波提取时间10 min,微波功率450 W,在该条件下石榴皮多糖的得率为10.48%。通过MTT法、中性红法、NO释放量测定和黏附试验初步表明石榴皮多糖对RAW264.7巨噬细胞具有一定的免疫调节作用。

Abstract: In this paper,the microwave assisted extraction of pomegranate peel polysaccharide was optimized and itsimmunomodulatory activity was studied.Based on the single factor experiment,the optimum extraction condition was determined by response surface analysis method.The solid-liquid ratio,extraction time,and microwave power were chosen as independent variables and the yield of pomegranate peel polysaccharide was selected as response value.Under the optimum extraction condition of solid-liquid ratio of 44∶1 mL/g,extraction time of 10 min,and microwave power of 450 W,the yield of pomegranate peel polysaccharide was 10.48%.In addition,the immunomodulatory effects of macrophage RAW264.7 was preliminarily verified by MTT,neutral red essay,NO release determination,and adhesion test.
