响应面法优化金银花中环烯醚萜苷的提取工艺及其产地差异

Optimization of Extraction for Iridoid Glycosides from Lonicerae Flos by Box-Behnken Response Surface Methodology and Differences in Iridoid Glycosides Content among Different Areas

摘要: 为优化金银花中环烯醚萜苷的超声提取工艺,以马钱苷酸、马钱苷与当药苷的提取率为指标,运用单因素试验结合Box-Behnken中心组合设计的方法进行提取工艺的优化研究。金银花中环烯醚萜苷的最佳提取工艺:液料比117 mL/g、乙醇浓度34%、超声时间45 min,在此条件下环烯醚萜苷的实际提取率为3.26 mg/g,与理论值3.28 mg/g的相对偏差为0.31%。同时发现金银花中环烯醚萜苷的含量在不同产地间存在明显差异。Box-Behnken响应面优化所得的金银花提取工艺稳定、可行,可较好的预测实验结果。

Abstract: To optimize process conditions of ultrasonic extraction of iridoid glycosides from Lonicerae Flos,with the extraction rates of loganic acid,loganin,sweroside as evaluation indexes,the single factor experiment combined with Box-Behnken design was used to optimize the extracting technology.The optimal extraction conditions were as follows:the liquid-to-solid ratio was 117 mL/g,and the ethanol concentration was 34%,the ultrasonic extraction time was 45 min,the predicted value of extraction rates was 3.28 mg/g.Under the extraction conditions after optimization,the value of extraction rates was 3.26 mg/g,and the relative deviation between the predicted and measured value was 0.31%.Meanwhile,there was great variation in iridoid glycosides content among different areas.The ultrasonic extraction technology process conditions of iridoid glycosides from Lonicerae Flos optimized by the response surface method,which was suitable and feasible.
