牛筋果中酚性化合物及其对DPPH自由基的清除活性

摘要: 本文对牛筋果Harrisonia perforata(Blanco) Merr.中的酚性成分及其抗氧化活性进行了研究。从牛筋果小枝中分离得到了14个酚性化合物,分别鉴定为:去甲丁香色原酮(1)、柴胡色原酮A(2)、5,7-dimethoxy-2-methylchromone(3)、2-hydroxymethylalloptaeroxylin(4)、羟苯乙酯(5)、苯丙酮(6)、 cephafortin B(7)、去氢双松柏醇(8)、hawthornnin C(9)、hawthornnin D(10)、丁香树脂酚(11)、皮树脂醇(12)、4-ketopinoresinol(13)、罗汉松脂酚(14)。化合物135~1012~13共10个化合物为首次从植物牛筋果中分离得到。使用DPPH法筛选了化合物的抗氧化活性,其中,化合物1314抗氧化活性优于阳性对照(其EC50值分别为53.88±5.34和32.67±3.96 μM),并对其构效关系进行了初步探讨。该结果为牛筋果的深度开发利用提供了理论依据。

Abstract: The study was focus on the phenolic compounds of Harrisonia perforata(Blanco) Merr.and their antioxidant activity.Fourteen compounds were isolated from the branches of Harrisonia perforata(Blanco) Merr.by various column chromatography.Their structures were identified as noreugenin (1),saikochromone A (2),5,7-dimethoxy-2-methylchromone (3),2-hydroxymethylalloptaeroxylin (4),ethylparaben (5),propiophenone (6),cephafortin B (7),dehydroconiferyl alcohol (8),hawthornnin C (9),hawthornnin D (10),syringaresinol (11),medioresinol (12),4-ketopinoresinol (13)and matairesinol (14)based on ESI-MS,1H and 13C NMR spectroscopic data.Among them,compound 1,3,5-10,and 12-13 were isolated from this plant for the first time.In addition,DPPH radical scavenging method was used to evaluate antioxidant activity,compound 13 and 14 showed strong DPPH free radical scavenging capacity.And then,we discussed the structure activity relationship of these phenolic compounds.The results provided a theoretical basis for deep development and utilization of Harrisonia perforata(Blanco) Merr.
