HPLC法分析不同年限及不同部位掌叶大黄9种成分的积累特征

Accumulation profile of nine constituents in different years old stage and parts of Rheum palmatum  L. by HPLC analysis

摘要: 为研究不同年限及部位掌叶大黄9种成分含量及其变化特征。利用HPLC法进行分析,色谱柱为武本C18 (5 μm,4.6 mm×250 mm),流动相为甲醇-磷酸水(0.2%),检测波长260 nm,柱温30 ℃,体积流量1.0 mL/min,进样量10 μL。线性范围良好(R2>0.995),精密度、稳定性、重复性RSD均小于2%,加样回收率97.30%~108.20%。含量分析表明:同一部位,随着年限增加,除根中没食子酸、根茎中大黄素、叶片中大黄酚的含量无变化外,其他8成分的含量均随年限增加而增加或者第4年增加、第5年无显著变化。同一年限,大黄酚-8-O-葡萄糖苷、大黄素、大黄酚、大黄素甲醚的含量次序为根>根茎>叶片;3年生芦荟大黄素,4年生没食子酸、番泻苷B叶片中含量与根中大致相等且高于根茎;5年生大黄素-8-O-葡萄糖苷的含量次序为叶片>根茎>根,根与根茎中没食子酸、大黄酚的含量相当且高于叶片。本实验成功建立简便快捷、重现性好的HPLC分析方法,明确了不同年限及部位掌叶大黄9种成分积累特征,为大黄质量评价和药材高效生产提供理论依据。

Abstract: To analyze the contents and variation rules of 9 components in different years old stage and parts of Rheum palmatum L.The HPLC analysis was performed on Woburn C18 chromatographic column (5 μm,4.6 mm×250 mm),the detection wavelength was set at 260 nm,the column temperature was 30 ℃,the flow rate was 1.0 mL/min,with the injection volume of 10 μL.The linear range was good (R2>0.995),RSDs of precision,stability,and repeatability tests were all low than 2%,and the average recoveries were 97.30%-108.20%.The contents analysis showed that,in the same part,except for the relative stability of gallic acid in the root,emodin in the rhizome,and chrysophanol in the leaf,the contents of the other components increased year by year or at the 4th year,and showed no significant change at the 5th year.During the same year,the contents of chrysophanol-8-O-glucoside,emodin,chrysophanol,physcion ether was in the order of root >rhizome > leaf.Contents of aloe emodin at the 3rd year,gallic acid,and sennoside B at the 4th year in leaves were as much as those in roots.The contents of emodin-8-O-glucoside at the 5th year was in the order of leaf > rhizome > root.The contents of gallic acid and chrysophanol in root and rhizome were the merely equal,however,higher than those in leaves,at the 5th year.The HPLC analysis method with simple,rapid,and good reproducibility was successfully established,clarifying the different accumulation characteristics of 9 components in different ages and parts of R.palmatum L.,providing a theoretical basis for its quality evaluation and efficient medicine production.
