赶黄草挥发油GC-MS分析及保肝靶点筛选

Analysis of chemical constituents and bioinformatics of Penthorum chinense

摘要: 为筛选赶黄草挥发油保肝作用的关键活性成分及靶标蛋白,本研究采用水蒸汽蒸馏法(steam distillation method)提取赶黄草中的挥发油,并结合GC-MS(gas chromatography-mass spectrometer) 进行分析鉴定,通过生物信息学对赶黄草挥发油主要化学成分(相对含量>2%)进行靶点筛选、生物功能和疾病分析,采用分子对接技术(sybyl 2.1.1)对关键活性成分进行验证,基于Ligplot1.4.5软件对活性成分与CNR1进行相互作用分析。结果从赶黄草挥发油中共检测出31种活性成分,靶点筛选得到6个hub靶点(CNR2、CNR1、CCR5、CCR3、PRKCA、PRKCB)。研究发现赶黄草挥发油可能通过调节大麻素受体2、大麻素受体1、C-C趋化因子受体5、C-C趋化因子受体3、蛋白激酶Cα、蛋白激酶Cβ防治肝硬化、肝炎和非酒精性脂肪肝等肝脏疾病。此外香叶基丙酮、2-硬脂酸单甘油酯和金合欢基丙酮为赶黄草挥发油保肝的关键活性成分。

Abstract: Screening the key active ingredients and target proteins with liver protecting effect from Penthorum chinense volatile oil.Extracting the volatile oils from Penthorum chinense by steam distillation method,and the analysis and identification were made by combing GC-MS.Based on bioinformatics,we made targets screening,biological function and disease analysis for the main chemical components of volatile oil (relative content > 2%) of Penthorum chinense.The key active ingredients were verified by molecular docking technology (Sybyl 2.1.1).Based on the software Ligplot1.4.5,we researched the interaction analysis between the active ingredients and CNR1.Thirty-one kinds of main active constituents of Penthorum chinense volatile oil were detected,and 6 hub targets (CNR2,CNR1,CCR5,CCR3,PRKCA,PRKCB) were included.The liver diseases such as cirrhosis,hepatitis and non-alcoholic fatty liver could be prevented and treated by regulating cannabinoid receptor 2,cannabinoid receptor 1,c-c chemokine receptor 5,c-c chemokine receptor 3,protein kinase Cα,and protein kinase Cβ with the help of using volatile oil of Penthorum chinense.Geranylacetone,2-monostearin and farnesylacetone were the key active ingredients of Penthorum chinense volatile oil.
