桦褐孔菌三萜提取纯化工艺及体外抗肿瘤作用研究

Study on extraction and purification of triterpenoids from inonotus obliquus and its anti-tumor activity

摘要: 为了优选桦褐孔菌三萜的提取工艺及大孔树脂优化工艺。本实验以总三萜提取率为指标,选择提取溶剂、料液比、提取温度、提取时间、提取次数为考察因素,通过单因素考察和响应面法筛选桦褐孔菌三萜提取工艺。选取上样浓度、上样体积、上样PH、上样流速、洗脱液浓度及体积为考察因素,采用单因素试验对AB-8型大孔树脂纯化最优工艺进行了研究。并通过MTT实验验证其抗胃癌活性。结果表明,用无水乙醇作为提取溶剂,90 HZ,超声30 min后,溶剂比1∶15,82 ℃提取2 h。最优桦褐孔菌三萜纯化工艺为上样浓度1 mg/mL,上样量2 BV,上样最佳pH=5,上样最佳流速2 mL/min,洗脱液选择95%乙醇,pH=5。采用纯化后的桦褐孔菌三萜进行体外MTT实验,结果得出桦褐孔菌三萜具有显著性抑制HeLa细胞生长作用。该提取及纯化工艺可行,为桦褐孔菌三萜的开发打下基础。

Abstract: In order to optimize extraction technology of Inonotus obliquus triterpenoids,and investigate its purification technology by macroporous resin.This experiment with the content of total saponins as index,extract solvent,solid-liquid ratio,extraction temperature,extraction time and times were selected as factors,the single-factor investigation and response surface method were used to screen the extraction process of Inonotus obliquus triterpenoids.With sample concentration,sample volume,flow rate on sample PH,concentration and volume of eluent were selected was used to investigate purification technology.The single-factor investigation was used to AB-8 macroporous resin purification process.The MTT test validates its anti-gastric activity.The results showed that ethanol was used as the extraction solvent,90 Hz,ultrasonic for 30 min,the solvent ratio was 1∶15 and extraction was 82 ℃ for 2 h.The optimal purification process for Inonotus obliquus triterpenes∶1 mg/mL for sample loading,2 BV for sample loading,optimal pH=5 for sample loading,2 mL/min for optimum flow rate,95% ethanol for eluent,pH=5.using purified Inonotus obliquus triterpenes can significantly inhibit HeLa cell growth.The extraction and purification process is feasible.And lay the foundation for the development of Inonotus obliquus triterpenoids.
