华泽兰花部位化学成分及其抑菌活性研究

Chemical constituents from flower of Eupatorium chinense and their antibacterial activity

摘要: 为研究菊科泽兰属植物华泽兰Eupatorium chinense L.花部位的化学成分,采用正相硅胶柱色谱、制备薄层色谱及制备型 HPLC 等方法分离纯化,利用 NMR、MS等波谱学方法鉴定化合物结构,从华泽兰花中得到了9个化合物,应用波谱学方法分别鉴定为:3,11-二氧,Δ12-齐墩果烯(1)、3β-(辛烷氧基)-11-氧代烯烃-12-烯-28-酸(2)、β-香树脂醇(3)、山柰酚-3-O-β-D-半乳糖苷(4)、咖啡酸乙酯(5)、5-甲基-2(1-甲基乙基)苯基-β-D-吡喃葡萄糖苷(6)、eupalinolide K(7)、11-羟基-10,11-二氢泽兰素(8)、dieupachinin E(9),其中化合物1~7均为首次从该种属植物中得到。化合物1~3为齐墩果烷型五环三萜,为华泽兰中首次分离得到,推测其为华泽兰花部分的特征成分。对化合物1~7进行了抗菌活性测试:采用牛津杯法评价部分化合物的抑菌活性,结果显示,化合物1~6显示出不同程度的抑菌活性,其中化合物14活性最好。本文是首次对华泽兰花部位的化学成分进行报道。

Abstract: To study chemical constituents of the flower of Eupatorium chinense,various chromatographic techniques,including normal phase silica gel column chromatography,preparative thin-layer chromatography,and preparative HPLC were used.Their structures were determined by various spectral data.Finally,nine compounds were isolated from the acetic ether extract of flower of E. chinense and the structures were identified as 3,11-dioxo-olean-12-ene(1),3β-(octanoyloxy)-11-oxoolean-12-en-28-oic acid(2),β-armyrin(3),kaempferol-3-O-β-D-galactoside(4),ethyl caffeate(5),5-Methyl-2-(1-methylethyl)phenyl-β-D-glucopyranoside(6),eupalinolide K(7),11-hydroxy-10,11-dihydro-euparin(8),dieupachinin E(9).Among them,compounds 1-7 were isolated from the flower of E. chinense for the first time.Compounds 1-3,belonging to oleanolic pentacyclic triterpenoids,were isolated for the first time from Eupatorium chinensis and presumed to be the characteristic constituent of the flower of Eupatorium chinensis. Moreover,the antimicrobial activity of compound 1-7 was tested and evaluated by Oxford cup method.The results showed that the tested compounds had different degrees of antimicrobial activity against bacteria or fungus.Compounds 1 and 4 had the best activity.The study was the first report about the chemical constituents of the flower parts of E. chinense.
