土茯苓降尿酸活性成分研究

Anti-uric acid constituents from Rhizoma Smilacis Glabrae

摘要: 研究土茯苓降尿酸功效成分。采用乙醇提取,多种层析分离,通过建立高血尿酸小鼠模型并结合黄嘌呤氧化酶体外抑制实验,筛选土茯苓降尿酸活性成分,利用核磁共振鉴定化学结构。结果表明,土茯苓醇提物大孔树脂分离得70%洗脱物能显著(P<0.01)降低小鼠血尿酸含量,70%洗脱物再经MCI、ODS柱分离,从该部分得到5个单体化合物,分别是落新妇苷(1)、花旗松素-3-O-α-L-吡喃鼠李糖基-α-L-吡喃鼠李糖苷(2)、黄杞苷(3)、异黄杞苷(4)、4-(吡咯并[3,2-d]嘧啶-4-基)苯酚(5)。化合物2~5均对黄嘌呤氧化酶的活性有显著抑制作用(P<0.01),化合物5的抑制作用最强,抑制率达到80.47%。

Abstract:

This research aims to investigate the active constituents of Rhizoma Smilacis Glabrae and its anti-uric acid effect on mice.The active constituents was extracted by ethanol reflux,separated and purified by multiple chromatography.The structure of the obtained product was identified by NMR.A mouse model with high blood uric acid was established and combined with xanthine oxidase in vitro inhibition experiment to screening the active constituents.Results showed that 70% ethanol eluate from the macroporous resin extracted from Rhizoma Smilacis Glabrae had the most obvious anti-uric acid effect on mice (P<0.01).After separation by MCI and ODS column,five compound monomers were further separated:astilbin (1),Taxifolin-3-O-α-L-rhamnopyranosyl-α-L-rhamnopyranoside (2),engeletin (3),isoengeletin (4),4-(pyrrolo[3,2-d]pyrimidin-4-yl)pheno (5).Compounds 2-5 had significant inhibitory effect on xanthine oxidase activity (P<0.01);Compound 5 showed the strongest inhibitory effect with an inhibitory rate of 80.47%.
