大叶糖胶树吲哚生物碱化学成分研究

Indole alkaloids from Alstonia macrophylla

摘要:

为研究大叶糖胶树(Alstonia macrophylla)化学成分及生物活性。采用正相硅胶、葡聚糖凝胶、制备液相等色谱技术,对大叶糖胶树中的化学成分进行了分离纯化,共分离得到6个化合物,通过NMR、MS等波谱方法将其结构鉴定为vallesamine(1)、picrinine(2)、khasuanine A(3)、scholaricine(4)、echitamine(5)、strictosamide(6)。化合物3和6首次从该属植物中分离得到,化合物1、2、4、5为首次从该植物中分离得到。所有化合物测试了对HCT-116人结肠癌细胞和DLD-1人结直肠腺癌上皮细胞的细胞毒性,结果显示化合物2和4具有一定的细胞毒性,化合物4对DLD-1人结直肠腺癌上皮细胞的细胞毒性较为明显(IC50 =16.7 μg/mL),其余化合物均未显示出生物活性。

Abstract:

In order to investigate the chemical constituents and biological activities of Alstonia macrophylla, six monoterpenoid indole alkaloids,namely vallesamine (1),picrinine (2),khasuanine A(3),scholaricine(4),echitamine(5),strictosamide(6) were isolated from leaves and twigs of Alstonia macrophylla by repeated column chromatography over silica gel,Sephadex G and Prep-HPLC.Their structures were elucidated based on NMR and MS spectroscopic data.Compounds 3 and 6 were firstly obtained from this genus,and compounds 1,2,4 and 5 were isolated from this plant for the first time.All compounds were evaluated the cytotoxicity of HCT-116 and DLD-1.Among them,compounds 2 and 4 had certain cytotoxicity,and compound 4 showed DLD-1 inhibitory activity with an IC50 value of 16.7 μg/mL.
