扶芳藤提取物体内体外抗氧化作用研究

摘要:

研究扶芳藤各极性萃取部位的体外及体内抗氧化作用,为扶芳藤的开发利用提供科学参考。以DPPH法及ABTS法考察扶芳藤各极性部位的体外抗氧化活性;以LPS(1g/L)刺激的小鼠急性肺炎为动物模型,测定血清中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性和丙二醛(MDA)含量,考察各极性部位的体内抗氧化活性。结果显示扶芳藤各极性部位对DPPH自由基都具有清除作用,正丁醇部位、乙酸乙酯部位及石油醚部位的IC50分别为0.103、0.258及0.702 mg/mL;扶芳藤各极性萃取部位对ABTS的产生也都具有抑制作用,正丁醇部位、乙酸乙酯部位及石油醚部位的IC50分别为0.814、2.384及2.059 mg/mL。体内实验结果表明,乙酸乙酯部位和正丁醇部位对改善小鼠体内SOD、MDA和GSH-Px活性有着良好的效果,且与模型组比较有显著性差异P<0.05。证明扶芳藤分离的各萃取极性部位在体内体外均具有不同程度的抗氧化效果,综合考虑以乙酸乙酯和正丁醇提取部位活性为最佳,石油醚部位则活性最差。

Abstract:

To investigate the in vivo and in vitro antioxidant effects of extract from Euonymus fortunei (Turcz.) Hand.-Mazz.The in vitro antioxidant activities were studied by DPPH and ABST radical scavenging.Acute lung injury mouse models induced by LPS were used to investigate the in vivo antioxidant activities by measuring SOD,GSH-Px activities and MDA content in serum.The different extraction parts of Euonymus fortunei(Turcz.)Hand.-Mazz had scavenging effects on DPPH.The IC50 values of n-butanol,ethyl acetate and petroleum part ether were 0.103,0.258 and 0.702 mg/mL,respectively.The different polar extraction parts of Euonymus fortunei(Turcz.)Hand.-Mazz had inhibitory effects on the production of ABTS.The IC50 values of n-butanol,ethyl acetate and petroleum part ether were 0.814,2.384 and 2.059 mg/mL,respectively.Comprehensive activity results in vivo,n-butanol,ethyl acetate and petroleum part ether had good effects on increasing SOD,MDA and GSH-Px activities,and there were significant differences compared with the positive control group.The various eparation components all have good antioxidant activities both in vitro and in vivo.Considering the indicators both in vitro and in vivo,the Ethyl acetate and n-butanol parts were the best,and the activity of the petroleum ether site was the worst.
