中华真地鳖醇提物对人前列腺癌PC3细胞体外作用及其机制研究

摘要:

探究中华真地鳖醇提物(ESWE)对人前列腺癌PC3细胞生长、迁移和侵袭的影响及作用机制。采用MTT法检测ESWE对PC3细胞的毒活性,流式细胞术、Hoechst 33258染色检测细胞凋亡情况,划痕实验和Transwell细胞侵袭实验检测ESWE对肿瘤细胞体外迁移和侵袭作用的影响,Western Blot 法测定不同浓度ESWE处理PC3细胞后,转移相关蛋白金属基质蛋白MMP-2和MMP-9的表达。结果表明,ESWE对人前列腺癌PC3细胞的生长、迁移和侵袭有明显的抑制作用,呈一定的剂量依赖关系,且能下调转移相关蛋白MMP-2和MMP-9的表达。流式和凋亡染色结果显示,ESWE不能诱导PC3细胞凋亡。综上说明ESWE能够抑制人前列腺癌PC3细胞的增殖、迁移和侵袭能力,其机制可能与下调MMP-2和MMP-9蛋白表达有关。

Abstract:

To investigate the effect of Eupolyphaga sinensis Walker ethanol extract (ESWE) on human prostate cancer PC3 cells growth,migration and invasion in vitro,and to study its mechanism.MTT assay was used to determine the toxicity of ESWE on human prostate cancer PC3 cells.Cell apoptosis were analyzed by flow cytometry and Hoechst 33258 staining.Transwell assay and cell scratch test were used to detect the effect of ESWE on migration and invasion of PC3 cells.Western blot assay were employed to detect the expressions of metastasis related proteins MMP-2 and MMP-9 after different concentrations of ESWE were used to treat human breast cancer.The results showed that ESWE can inhibit the growth,migration and invasion of human prostate cancer PC3 cells in a concentration-dependent manner.Furthermore,ESWE could reduce the protein expression of MMP-2 and MMP-9.The results of flow cytometry and Hoechst 33258 staining showed that ESWE could not induce PC3 cell apoptosis In conclusion,ESWE can inhibit the proliferation,migration,invasion of human prostate cancer cell line PC3,and the mechanism may be associated with the inhibition of the expression of MMP-2 and MMP-9.
