白及须根多酚提取工艺优化及其抗氧化、酪氨酸酶抑制活性研究

摘要:

通过响应面法优化白及须根多酚(polyphenols from fibrous roots of Bletilla striata,BRP)的提取工艺,并评价BRP的体外抗氧化、酪氨酸酶抑制活性。以多酚含量为响应值,采用绿色溶剂甘油联合高剪切分散乳化技术(high-shear mixing emulsification technique,HSME)提取BRP,在单因素实验的基础上,采用响应面分析法优化提取工艺参数。以ABTS与DPPH自由基清除能力、还原能力和抑制酪氨酸酶作用为评价指标,研究BRP体外抗氧化、酪氨酸酶抑制活性。结果显示,在剪切时间150 s,料液比1∶50(g/mL)的条件下,当甘油浓度为53%,pH为4,剪切转速为16 200 rpm,剪切温度为44 ℃时,BRP 提取量最高,为19.02 mg/g。BRP具有良好的DPPH、ABTS自由基清除能力和还原能力,其抗氧化活性与浓度呈显著正相关(P<0.05)。BRP和Vc均能显著抑制酪氨酸酶活性,其酪氨酸酶的半数抑制浓度(half-inhibition concentration,IC50)值分别为0.124和0.287 mg/mL。本研究为白及须根的合理开发利用提供了理论基础。

Abstract:

This study was conducted to optimize the extraction process of polyphenols from fibrous roots of Bletilla striata (BRP) by response surface methodology and investigate its antioxidant and tyrosinase inhibitory activities in vitro.The method of high-shear mixing emulsification technique (HSME) combined with the green solvent glycerol was used,and on the basis of single-factor test,the extraction process of BRP was optimized by response surface analysis.The antioxidant activity of BRP was examined by reducing power,DPPH and ABTS free radical scavenging assays.Additionally,the tyrosinase inhibitory activity of BRP was explored.The results showed that the optimal process conditions were as follows,shear time of 150 s,solid-liquid ration of 1∶50 g/mL,glycerol concentration of 53%,pH value of 4,shear speed of 16 200 rpm and temperature of 40 ℃.Under these conditions,the content of BRP was 19.12 mg/g.BRP exhibited strong reducing power,DPPH and ABTS free radicals scavenging abilities,and the antioxidant activity demonstrated significant correlations with the content of BRP (P<0.05).Both BRP and Vc exerted potent tyrosinase inhibitory capacities with the IC50 values of 0.124 and 0.287 mg/mL,respectively.This research can provide a theoretical basis for the rational development and comprehensive utilization of fibrous roots of Bletilla striata.
