响应面法优化酶解碱提沙棘籽粕蛋白的工艺研究

Optimization of protein extraction process from Hippophae rhamnoides seed meal by response surface methodology

摘要: 本文以沙棘籽粕为原料,采用醇提法预处理材料,以酶解碱提联用法提取蛋白粗提物,通过单因素实验,对影响沙棘籽粕蛋白提取得率的液料比、温度、pH、胰蛋白酶加酶量、提取时间、提取次数等因素进行筛选和初步优化,通过基于Box-Behnken Design的响应面法研究最佳提取工艺,验证响应面优化结果。分析响应面结果,沙棘籽粕蛋白提取的最优工艺参数为:液料比44∶1(mL∶g)、pH12.3、胰蛋白酶加酶量1.4%、温度60 ℃、提取时间30 min、提取次数3次。此条件下,蛋白实际提取得率50.53%,接近数学模型的预测值51.54%。

Abstract:

In this study,the protein was extracted by enzymatic hydrolysis alkali solubilization from the Hippophae rhamnoides seed meal prepared by alcohol extraction.Through the single factor experiments,the factors affecting the protein extraction yield were screened and preliminaries optimized.The optimum conditions of liquid-to-solid ratio,pH and trypsin amount were obtained via response surface of Box-Behnken design.Experimental results showed the optimal protein extraction conditions are liquid-to-solid ratio 44∶1(mL∶g),temperature 60 ℃,pH12.3,trypsin amount 1.4%,extraction time 30 min and 3 times.The validation experiment demonstrated optimization results.Under the conditions,the actual yield of protein was 50.53%,closed to predicted value 51.54% by model.
