毛叶藜芦中甾体生物碱鉴定及其促进溶酶体生成活性的研究

摘要:

研究毛叶藜芦中的甾体生物碱类成分及其促进溶酶体生成活性。应用柱层析,反相色谱,凝胶色谱等手段对毛叶藜芦根部的甲醇提取物进行分离,再根据其理化性质、UHPLC-MS/MS、NMR等现代波谱技术,对分离得到的生物碱进行结构鉴定,并通过Red DND-99溶酶体红色荧光探针对甾体生物碱促进溶酶体生成活性进行初步筛选研究。鉴定了8种甾体生物碱,分别为veratramine(1)、solasodine (2)、epirubijervine(3)、jervine(4)、3-acetyl-15-methylbutyroylgermine(5)、isorubijervine(6)、23-methoxy-cyclopamine(7)、rubijervone-12(8),其中23-methoxy-cyclopamine(7)和rubijervone-12(8)首次从毛叶藜芦中分离得到,化合物6可以明显促进溶酶体生成;溶酶体对于维持神经系统的稳态非常重要,与神经退行性疾病紧密相关,探究甾体生物碱对溶酶体的生成作用,为神经退行性疾病的治疗提供先导化合物。

Abstract:

To study the steroidal alkaloids of the roots and rhizomes of Veratrum grandiflorum Loes. and their activities of promoting lysosomal biosynthesis.The methanol extracts from the roots of V. grandiflorum were separated by column chromatography,reversed-phase chromatography and gel chromatography.The structures of the isolated alkaloids were identified by the physical and chemical properties,UHPLC-MS/MS,NMR and other spectroscopic techniques.The activities of lysosome biosynthesis were preliminarily screened by Red DND-99 lysosome red fluorescence probe.Eight steroidal alkaloids were isolated and their structures were identified as veratramine (1),solasodine (2),epirubijervine (3),jervine (4),3-acetyl-15-methylbutyroylgermine (5),isorubijervine (6),23-methoxy-cyclopamine (7) and rubijervone-12 (8).23-Methoxy-cyclopamine (7) and rubijervone-12 (8) were isolated from this plant for the first time.Compound 6 promoted lysosome biosynthesis obviously;Lysosomes are very important for maintaining the homeostasis of the nervous system and are closely related to neurodegenerative diseases.To explore the effect of steroid alkaloids on the lysosome biosynthesis,and will provide lead compounds for the treatment of neurodegenerative diseases.
