化学修饰L02肝细胞膜生物亲和材料快速筛选大黄降血脂活性成分

Rapid screening of lipid-lowering active constituents from Rheum palmatum L.with chemically modified L02 liver membrane bioaffinity material

摘要: 为增加细胞膜生物亲和材料使用寿命,建立以APTES修饰硅胶为载体的L02细胞膜生物亲和材料,并将其应用于大黄降血脂活性成分的快速筛选。采用油酸刺激L02肝细胞,建立肝脂肪变性模型;硅胶与APTES、戊二醛发生反应,在硅胶表面引入醛基,游离醛基与细胞膜上富含的氨基通过共价键连接;扫描电镜和红外光谱进行表征;对大黄30%乙醇提取液进行吸附,HPLC分析吸附结果。通过扫描电镜证实材料表面覆盖有一层细胞膜,红外光谱也出现3 442、2 942 cm-1的-NH 键特征吸收峰,表明材料制备成功;HPLC分析表明:11种化学成分被特异性吸附,分别为:芦荟大黄素-8-O-β-D-葡萄糖苷、大黄酸-8-O-β-D-葡萄糖苷、大黄酚-8-O-β-D-葡萄糖苷、大黄素甲醚-8-O-β-D-葡萄糖苷、芦荟大黄素、大黄酸、大黄素、大黄酚、大黄素甲醚及2种未知成分。制备的化学修饰L02肝细胞膜生物亲和材料能够延长使用次数,也可用于大黄及其他中药降血脂活性成分的快速筛选。

Abstract:

To extend life span of cell membrane material,a novel type of 3-aminopropyltriethoxysilane (APTES) decorated silica gel was employed. A L02 hepatic steatosis cell model was established using oleic acid.Silica gel was decorated with glutaraldehyde with the help of APTES,which react with the amino groups on L02 hepatic steatosis cell membranes to form a covalent bond.For the material characterization was used Scanning electron microscopy (SEM) and Fourier transform infrared spectroscopy (FTIR).The material was selected as an adsorptive material to screen for the potential active ingredients of Rheum palmatum L.by HPLC.SEM confirmed that the surface of silica gel was obviously covered with cell membrane.The characteristic absorption peaks of -NH bond were 3 442 and 2 942 cm-1,indicating that the material was successfully prepared.HPLC analysis showed that 11 chemical components were specifically adsorbed,which were identified as aloe-emodin-8-O-β-D-glucopyranoside,rhein-8-O-β-D-glucopyranoside,emodin-8-O-β-D-glucoside,physcion-8-O-β-D-monoglucoside, aloe-emodin, rhein,emodin,chrysophanol,physcion and two unknown components.The chemically modified L02 liver membrane bioaffinity material can prolong its life span and use for rapid screening of the lipid-lowering active ingredients of Rheum palmatum L.and other traditional Chinese medicine.
