裂果薯皂苷的特征图谱及总皂苷中有效组分的抗肿瘤活性研究

The Characteristic Spectra of Saponins of Schizocapsa Plantaginea Hance and the Antitumor Activity of Effective Components in the Total Saponins

摘要: 采用高效液相色谱紫外及蒸发光散射检测器建立裂果薯总皂苷(SFSP60%)的特征图谱,乙腈-水为流动相梯度洗脱,检测波长203 nm,柱温30 ℃,流速1 mL/min,进样量10 μL。以ELSD色谱图中SFSP60%色谱峰为参照,采用面积归一法计算出裂果薯皂苷Ⅰ(SPHSⅠ)相对百分比为84.28%,裂果薯皂苷Ⅱ(SPHSⅡ)为302%,化合物1为2.21%,化合物2为6.31%,化合物3为2.73%,化合物1~3可能是母核类似的皂苷类化合物。取对数生长期的人肝癌SMMC-7721细胞,给药组分别加入0、0.25、0.5、2、4 μg/mL的SFSP60%、SPHSⅠ、SPHSⅡ和SPHSⅠ+Ⅱ(SPHSⅠ和SPHSⅡ各占比96.54%、3.46%)培养24 h、48 h、72 h,采用MTT法观察并计算细胞增殖抑制率和IC50。结果显示给药组对SMMC-7721细胞的增殖抑制作用呈现时间与剂量依赖性(P均<0.05),且活性大小:SFSP60%< SPHSⅠ、SPHSⅡ < SPHSⅠ+Ⅱ。证明在总皂苷中是SPHSⅠ和SPHSⅡ发挥主要抗肿瘤作用,且抗肿瘤作用是各组分共同作用的结果。为裂果薯中有效组分的抗肿瘤活性研究提供了参考以及有效组分的合理配比提供了新思路。

Abstract: The characteristic spectra of total saponins of Schizocapsa plantaginea Hance (SFSP60%) were established by High Performance Liquid Chromatography with Ultraviolet and Evaporative Light Scattering Detector.Acetonitrile-water mobile phase gradient Elution.The detection wavelength was 203 nm,the column temperature was 30 ℃ and the flow rate was 1 mL/min.Sample volume 10 μL.Reference to the SFSP60% chromatographic peaks in the ELSD chromatogram.Area normalization method is used to calculate saponinⅠof Schizocapsa plantaginea Hance(SPHSⅠ) accounted for 8428%(The relative percentage),the saponinⅡ of Schizocapsa plantaginea Hance (SPHSⅡ) was 3.02%,the compound 1 was 2.21%,the compound 2 was 6.31%,the compound 3 was 2.73%,Compounds 1-3 may be nucleosomal similar saponins.Take the logarithmic growth phase SMMC-7721 cells,cells in the administration group were added to 0,0.25,0.5,2,4 μg/mL SFSP60%,SPHS Ⅰ,SPHS Ⅱ and SPHS Ⅰ+Ⅱ (SPHS Ⅰ and SPHS Ⅱ accounted for 96.54%,3.46%) for 24,48,72 h.MTT assay was used to detect the proliferation inhibition rate of SMMC-7721 cells in order to finally calculate its half inhibitory concentration (IC50).The results showed that the inhibitory effect of the administration group on the proliferation of SMMC-7721 cells was in a time-and dose-dependent manner (all P<0.05),and the viability size was SFSP60% < SPHSⅠ,SPHSⅡ < SPHSⅠ+Ⅱ.It was proved that SPHSⅠand SPHSⅡplay the main antitumor effect in total saponins,and the anti-tumor effect is the result of the interaction of each component.Which provided a reference for the study of the viability of effective components in Schizocapsa plantaginea Hance and a new idea for the rational allocation of effective components.
