Study on the anti-inflammatory activity of euptox A and 9-oxoageraphorone in Eupatorium adenophorum Spreng#br#

Abstract:

In this study,the in vitro anti-inflammatory activities of several extracts or purified monomers from Eupatorium adenophorum Spreng were investigated in order to find the major anti-inflammatory active ingredients of E. adenophorum.The extracts were obtained using supercritical fluid extraction CO2-(SFE),supercritical CO2 extraction-molecular distillation-(MDE) and methanol extract-(MSE),separately,and the tested monomers were euptox A and 9-oxoageraphorone.In vitro lipopolysaccharide (LPS)-induced RAW264.7 inflammation model of mouse mononuclear macrophages was used and NO and TNF-α inflammatory factors were measured to study the anti-inflammatory activities.All extracts and monomers showed well anti-inflammatory activities,but the inflammatory activity of the two monomers were higher than those of extracts.The NO inhibition rate of 9-oxoageraphorone (IC50=10.09 μg/mL) was higher than that of euptox A (IC50 = 16.23 μg/mL).It was found that extracts rich in euptox A and 9-oxoageraphorone had good anti-inflammatory activities,as well as the purified monomers,indicating that euptox A and 9-oxoageraphorone were the effective anti-inflammatory components of E. adenophorum.This article provided a basis for the resource development and utilization of E. adenophorum.
