不同种源的霍山石斛及霍山产铁皮石斛、铜皮石斛化学成分GC-MS分析

GC-MS Analysis of Chemical Components in Different Origins ofDendrobium huoshanense,Dendrobium henanense as well as Dendrobium  officinale and Dendrobium moniliforme Originated from Huoshan County

摘要: 研究不同种源的霍山石斛及霍山产铁皮石斛、铜皮石斛化学成分,为石斛资源保护和利用提供依据。石斛鲜品冷冻干燥至恒重后甲醇超声提取1.0 h,滤液冷冻干燥除去甲醇后溶于正己烷,气相色谱质谱联用法分析萃取液组分,质谱相似度、保留指数比对及标准品比对法鉴鉴定分离组分、半定量法测定分离组分相对含量,主成分分析法确定6种石斛样品主要差异成分。从六种供试品中分离出109种组分,鉴定出72种成分,主要为烷烃、酯、醇、酚、萜及甾体类成分;不同种源的霍山石斛及霍山产铁皮石斛、铜皮石斛中化合物种类和含量存在差异。主成分分析显示,化合物7(3甲基癸烷)、9(2,4二甲基正十一烷)、15(2,7,10三甲基十二烷)、17(4丙基苯酚)、37(4甲氧基桂皮酸甲醛)、43(1十三醇)、46(3,4二甲氧基5羟基苯甲醛)57(十五烷酸)、62(1十九烯)、6 5(十八烷)、672(二十烷)、88(十三烷二酸二乙酯)、91(橄榄油醇)、96(二十四烷)、98(蓖麻油酸)、101(2,2′亚甲基双(4甲基6叔丁基苯酚))、106(3β羟基5胆稀酸),为六种供试品的主要特征性成分,可用来区分六种不同的石斛样品。不同种源的霍山石斛及霍山产铁皮石斛、铜皮石斛化学成分存在差异。研究结果可为石斛资源开发、质量标准制定及质量控制提供科学根据。

Abstract: To research the difference of chemical components in different origins of Dendrobium huoshanense C.Z.Tang et S.J.Cheng,Dendrobium officinale Kimura et Migo and Dendrobium moniliforme (Linn.) Sw.and to provide references for protection and utilization of Dendrobium plants.The fresh collected Dendrobium samples were washed thoroughly in water and then freezedried by a MicroModulyo lyophilizer,powdered in a blender and extracted with ultrasonic by methanol for 1.0 h.Then the filtrate was freezedried to remove the methanol and the residues was redissolved in water and extracted by nhexane.The nhexane extraction was remove for further GCMS analysis.The isolated chemicals were identified by comparing their RM and KI values and the known chemical in the NIST2010 library.The quantitative analysis of each component can be directly calculated by the overall volume integration.At last,a PCA analysis was applied to investigate the main variation of chemical in the six Dendrobium samples.Total 109 chemicals were isolated and 72 of them were identified.The main compositions in the six Dendrobiums were alkane,ester,alcohols,phenol,terpenes and sterols.The compositions and contents were different in different origins of D.huoshanense,D.officinale and D.moniliforme.The PCA analysis resulted that the first three principal components explained 92.3% of the variance of the data and the loading plot of the three first components showed a strong relationship between seventeen major compounds 7、9、15、17、37、43、46、57、65、72、80、88、91、96、98、101、106.The chemical compositions were different among different origins of D.huoshanense,D.officinale and D.moniliforme.The research provides a scientific basis for the further development and utilization of Dendrobium resources.
