顶空固相微萃取-气质联用法测定白术不同部位挥发性成分

摘要:

本文主要是通过对白术主根、须根、茎、叶四个部位的挥发性成分进行测定和定性定量分析,获得其主要挥发性成分的组成和含量差异。研究方法是采用顶空固相微萃取-气质联用技术(HS-SPME-GC-MS)测定白术样品中挥发性成分,通过比对标准品、NIST谱库检索、结合文献对其挥发性成分进行鉴定,并用峰面积归一法测定各组分的相对含量。结果显示,在白术不同部位中初步鉴定出70种成分,通过对比标准品溶液确定了苍术酮这一关键成分;从主根部位共鉴定27种挥发性成分,苍术酮(37.79%)为主要成分;须根部位共鉴定35种挥发性成分,主要为苍术酮(25.85%)、β-倍半水芹烯(11.68%)、瓦伦亚烯(9.47%)等;茎中共鉴定32种挥发性成分,β-倍半水芹烯(12.09%)、苍术酮(11.54%)含量相对较高;叶中共鉴定45种,主要成分为大根香叶烯D(18.74%)、D-柠檬烯(12.89%)、苍术酮(5.98%)等。分析白术不同部位样品挥发性成分发现,苍术酮在白术非药用部位亦有较高含量;白术叶中挥发性成分组成丰富,萜烯类化合物是其主要的挥发性成分,在医药、食品、农业等领域具有潜在较高开发利用价值。本实验为白术不同部位挥发性成分综合利用奠定了基础。

Abstract:

This research mainly focuses on the determination of the volatile components in different parts of Atractylodes macrocephala Koidz. and carries out qualitative and quantitative analysis,which can help the changing trend pursue the main volatile components.The volatile components in A. macrocephala samples were determined by headspace solid-phase microextraction-gas chromatography-mass spectrometry (HS-SPME-GC-MS).The volatile components were identified by comparing standard samples,the NIST database,and literature.The peak area normalization method determined and compared each component's relative contents.A total of 70 components were preliminarily identified in A. macrocephala.In this paper,the composition and content differences of the main volatile components were obtained through the determination,qualitative and quantitative analysis of the volatile components in the main root,fibrous root,stem,and leaf of A. macrocephala. 27 volatile components were identified from the main root,and atractylodes (37.79%) was the main constituent.A total of 35 volatile components were identified in the fibrous root,mainly atractylon (25.85%),β-sesquiphene (11.68%),and valentene (9.47%),and so on.A total of 32 volatile components were identified in the stem,the contents of β-sesquifolene (12.09%) and atractylon (11.54%) were relatively high,and 45 species were identified in leaves.The main components were caryophyllene D (18.74%),D-limonene (12.89%),atractylon(5.98%),and so on.Based on the analysis of the volatile components in different parts of A. macrocephala,it was found that A. macrocephala has a high content in the non-medicinal parts of A. macrocephala,and the volatile components in the leaves of A. macrocephala are rich,and terpenes are the main volatile components.It has potential high development and utilization value in medicine,food,agriculture,and other fields.This experiment laid a foundation for comprehensively utilizing volatile components in different parts of A. macrocephala.
