淫羊藿苷对小鼠黑色素瘤B16细胞增殖、凋亡及迁移的影响

Effects of Icariin on Proliferation,Apoptosis and Migration of Mouse Melanoma B16 Cells

摘要: 观察淫羊藿苷对小鼠黑色素瘤B16细胞增殖、凋亡及迁移的影响。B16细胞经不同浓度的淫羊藿苷处理后,采用MTT法检测其对B16细胞增殖的影响,Hoechst 33258染色法观察B16细胞形态的变化,Annexin V-FITC/PI双染法检测B16细胞凋亡率,Western blot检测细胞内Bax,Bcl-2和Caspase-3蛋白表达的改变。同时,细胞划痕实验及Transwell小室迁移实验检测淫羊藿苷对B16细胞迁移能力的影响。与对照组相比较,淫羊藿苷能明显抑制B16细胞的增殖,且具有浓度依赖性。B16细胞形态由树突状变为固缩圆球状,染色质凝集,细胞凋亡率升高,相关蛋白Bax和Caspase3的表达升高,Bcl2的表达降低。另外,淫羊藿苷对B16细胞的迁移能力也具有明显抑制作用,并呈现浓度依赖性。由此可知淫羊藿苷能有效抑制B16细胞的增殖,诱导细胞凋亡,降低细胞迁移能力。

Abstract: To explore the effects of icariin on cell proliferation,apoptosis and migration of mouse melanoma B16 cells.After treatment with different concentrations of icariin,B16 cell proliferation inhibition rate was detected by MTT assay.The morphologic changes in B16 cells was observed after Hoechst 33258 staining.Cell apoptosis was evaluated by Annexin V-FITC/PI double staining.The protein levels of Bax,Bcl-2 and Caspase-3 in icariin-treated B16 cells were examined by western blot.Moreover,cell migration activity of icariin-treated B16 cells was examined by scratch wound assay and transwell migration assay.Compared with the control group,icariin significantly inhibited the proliferation of B16 cells in a concentration manner.Morphological changes including chromatic agglutination and nuclear fragmentation were observed in icariin-treated B16 cells.Meanwhile,the apoptosis rate of B16 cells was remarkably increased after icariin treatment.Western blot analysis showed that icariin up-regulated the protein levels of Bax and Caspase-3 and down-regulated the Bcl-2 protein level.In addition,icariin inhibited the migration of B16 cells in a concentration-dependent manner.Therefore,icariin can effectively inhibit cell proliferation,induce cell apoptosis and inhibit cell migration in B16 cells.
