圆齿野鸦椿果皮成分及抑菌活性研究

Study on the antifungal constituents from the pericarp of Euscaphis konishii Hayata

摘要: 为探究圆齿野鸦椿(Euscaphis konishii Hayata)果皮提取物对瓜果霉腐菌的抑制作用,本研究依据生物活性追踪法,并结合多种色谱分离技术与波谱鉴定手段从活性部位中分离鉴定18个化合物,分别为(7S,8R)﹣二氢脱氢二松柏醇9-O-D-吡喃葡萄糖苷(1)、楝叶吴萸素B(2)、(7R,8R)-threo-7,9,9'-三羟基-3,3'-二甲氧基-8-O-4'-新木脂素4-O-β-D-吡喃葡萄糖苷(3)、illiciumlignan B(4)、异鼠李素(5)、山奈酚(6)、槲皮素(7)、槲皮素-3-O-β-D-葡萄糖苷(8)、异鼠李素3-O-β-D-芸香糖苷(9)、夏佛托苷(10)、木樨草素-6-C-β-D-葡萄糖苷-8-C-α-吡喃阿拉伯糖苷(11)、对羟基苯甲醛(12)、香草酸(13)、[R-(R*,R*)]-(1R)-3-[(4R)-4-羟基-2,6,6-三甲基-1-环己烯-1-基]-1-甲基丙基-β-D吡喃葡萄糖苷(14)、(3E)-4-[(3R,4R,5R)-4-(β-D-吡喃葡糖氧基)-3,5-二羟基-2,6,6-三甲基-1-环己烯-1-基]-3-丁烯-2-酮(15)、长春花苷(16)、1-(4-乙基苯基)-1,2-乙二醇(17)、黑麦草内酯(18)。其中,除化合物6~812~13外,均为首次从该属植物中分离得到。此外,化合物1、9、10、11对瓜果霉腐菌具有较好的抑制作用,其EC50在56.06~123.94 μM之间。其中,化合物9对瓜果霉腐菌菌丝抑制的EC50值为56.06 μM,与阳性对照药丁子香酚活性相当。本研究首次发现圆齿野鸦椿果皮提取物对瓜果霉腐菌具有较好的抑制作用,并说明了黄酮碳苷类化合物是该植物的重要成分。

Abstract: In order to study the inhibitory effect of the pericarp extract of Euscaphis konishii Hayata on Pythium aphanidermatum.In this paper,based on the biological activity tracking method,a variety of chromatographic separation techniques and spectral identification analysis,18 compounds were isolated from E. konishii and identified as (7S, 8R)-dihdrodehydrodico-niferylalcohol4-O-β-D-glucopyranoside (1), evofolin-B (2), (7R,8R)-threo-7, 9, 9'-trihydroxy-3, 3'-dimethoxy-8-O-4'-neolignan4-O-β-D-glucopyranoside (3), illiciumlignan B (4), isorhamnetin (5), kaempferol (6), quercetin (7), quercetin-3-O-β-D-glucoside (8), isorhamnetin-3-O-β-D-rutinoside (9), xiafotoside (10), hibiscus-6-C-β-D-glucoside-8-C-α-pyranosin (11), 4-hydroxybenzaldehyde (12), vanillicacid (13), [R-(R*, R*)]-(1R)-[(4R)-4-hydroxy-2, 6, 6- trimethyl-1-cyclohexen-1-yl]-1-methylpropyl-β-D-glucopyranoside (14), (3E)-4-[(3R, 4R, 5R)-4-(β-D- glucopyranosyloxy)-3, 5-dihydroxy-2, 6, 6-trimethyl-1-cyclohexylen-1-yl]-3-buten-2-tone (15), roseoside (16), 1-(4-ethylphenyl)-1, 2-ethanediol (17), lolioide (18).Except for compounds 6-8,12-13,all others were isolated for the first time in this genus.Furthermore,the EC50 of compounds 1,9-11 against P. aphanidermatum were between 56.06-123.94 μM.The EC50 value of compound 9 against P. aphanidermatum was 56.06 μM,which was equivalent to the activity of the positive control drug eugenol.In this study,it was found for the first time that the extract of E. konishii had a good inhibitory effect on P. aphanidermatum,which indicated that the flavonoid glycosides were the important components of this plant.
