花椒中的异丁基酰胺化合物及其对HT22海马神经元铁死亡的抑制作用研究

Study on isobutyl amide compounds from Zanthoxylum bungeanum and their activities against ferroptosis in HT22 hippocampal cells

摘要: 对四川泸定县花椒的化学成分进行研究,以期寻找新的先导化合物。采用硅胶柱色谱、高效液相色谱等方法从花椒的70%盐酸水提取物中分离并鉴定了8个异丁基酰胺类化合物。通过高分辨质谱、核磁共振谱与文献对照,它们被鉴定为花椒素A(1)、hydroxy-α-sanshool(2)、tetrahydrobungeanool(3)、hydroxy-γ-isosanshool(4)、hydroxy-γ-sanshool(5)、bungeanool(6)、γ-sanshool(7)、isobungeanool(8)和β-谷甾醇(9)。化合物1为首次从该植物中分离得到。化合物3对erastin诱导的小鼠海马神经元细胞系HT22细胞铁死亡具有明显的抑制作用,IC50为4.08 μM。

Abstract: The chemical constituents of anthoxylum bungeanum collected in Luding County of Sichuan Province were investigated in order to develop new lead compound.Eight isobutyl amide compounds were isolated and identified from 70% hydrochloric acid extract of Z. bungeanum by silica gel column chromatography and high performance liquid chromatography.They were elucidated as zanthoxylumol A(1),hydroxy-α-sanshool(2),tetrahydrobungeanool(3),hydroxy-γ-isosanshool(4),hydroxy-γ-sanshool(5),bungeanool(6), γ-sanshool(7),isobungeol(8),and β-sitosterol(9) by high resolution mass spectrometry and nuclear magnetic resonance spectroscopy.Compound 1 was isolated from this plant for the first time.Compound 3 demonstrated good inhibitory effect on the hippocampal neuron cell line HT22 erastin-induced ferroptosis with IC50 to be 4.08 μM.
