千里香中黄酮类成分的分离鉴定及抗氧化活性研究

Isolation,identification and antioxidant activity of the flavones from Murraya panaculata (L.) Jack

摘要:

采用乙酸乙酯提取千里香的化学成分,以硅胶柱色谱法,结合制备色谱法和重结晶法等技术手段进行分离纯化,得到6个化合物,通过核磁共振波谱及文献数据分别鉴定为5,7,8,3′,4′-五甲氧基黄酮(1)、5,7,8,3′,4′,5′-六甲氧基黄酮(2)、5,7,3′,4′,5′-五甲氧基黄酮(3)、5,6,7,3′,4′,5′-六甲氧基黄酮(4)、5-羟基-6,7,3′,4′-四甲氧基黄酮(5)和5-羟基-6,7,8,3′,4′-五甲氧基黄酮(6)。化合物1为首次从九里香属植物中分离得到,同时也是首次从千里香植物中分离得到。清除DPPH自由基实验结果显示化合物1~4抗氧化活性很弱,含有5位羟基的化合物5、6抗氧化活性相对较强。

Abstract:

The plant of Murraya panaculata (L.) Jack was powered,extracted with ethyl acetate.The ethyl acetate extract was separated by silica gel column chromatography,preparative high performance liquid chromatography (HPLC) and recrystallization methods,then six compounds have been isolated.On the basis of nuclear magnetic resonance (NMR) and comparison of their spectra with values reported in the literature,the compounds designated as 5,7,8,3′,4′-pentamethoxyflavone (1),5,7,8,3′,4′,5′-hexamethoxyflavone (2),5,7,3′,4′,5′- pentamethoxyflavone (3),5,6,7,3′,4′,5′-hexamethoxyflavone (4),5-hydroxy-6,7,3′,4′-tetramethoxyflavone (5) and 5-hydroxy-6,7,8,3′,4′-pentamethoxyflavone (6).Compound 1 was isolated from the Murraya species and M. panaculata for the first time.The DPPH radical scavenging experiments of compounds showed that all of six compounds had antioxidant activity,among which 5-hydroxy-6,7,3′,4′-tetramethoxyflavone (5) and 5-hydroxy-6,7,8,3′,4′-pentamethoxyflavone (6) had stonger antioxidant activity.
