粗根荨麻果胶多糖的结构表征及免疫调节活性研究

Structural characterization and immunomodulatory activity of pectin polysaccharide from Urtica macrorhiza Hand.-Mazz.

本论文旨在研究从云南粗根荨麻(Urtica macrorrhiza Hand.-Mazz.)中提取出的一种新型均一多糖(UMRA),对其结构进行表征,并且初步探究其免疫活性。通过水提醇沉、季铵盐沉淀、凝胶柱层析等方法对粗根荨麻粗多糖进行纯化;经高效凝胶渗透色谱、紫外光谱、傅里叶变换红外光谱、甲基化以及核磁共振波谱等分析方法对其纯度及化学结构进行表征,并通过考察其对巨噬细胞活力及细胞因子分泌的影响,初步评价其免疫调节活性。研究结果表明,UMRA是一种重均分子量约为37.66 kDa的均一酸性多糖,主要由鼠李糖、半乳糖醛酸、半乳糖、葡萄糖醛酸、阿拉伯糖、甘露糖6种单糖组成,摩尔比为3.94∶3.34∶2.26∶1.00∶0.95∶0.46。结构分析表明其是一种具有I型鼠李半乳糖醛酸聚糖型骨架的果胶,以重复的[→2)-α-L-Rhap-(1→4)-α-D-GalpA-(1→]二糖单元为主链,半乳糖、甘露糖、阿拉伯糖和葡萄糖醛酸为侧链。UMRA在0.4~250 μg/mL浓度范围内对巨噬细胞无细胞毒性,能够促进巨噬细胞增殖并分泌一氧化氮(nitric oxide,NO)、肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)和白细胞介素-6(interleukin-6,IL-6)。本研究为云南粗根荨麻多糖的结构阐明以及进一步的药用研发奠定了基础。

Abstract:

This study aims to investigate a novel homogeneous polysaccharide (UMRA) extracted from Urtica macrorrhiza Hand.-Mazz. in Yunnan, China, focusing on characterizing its primary structure and preliminarily exploring its immunological activity. UMRA was purified through a series of steps, including water extraction, alcohol precipitation, quaternary ammonium salt precipitation, and gel column chromatography. Its purity and chemical structure were analyzed using high-performance gel permeation chromatography, ultraviolet spectroscopy, fourier-transform infrared spectroscopy, methylation, and nuclear magnetic resonance spectroscopy. The immunomodulatory activity of UMRA was evaluated by assessing its effects on macrophage viability and cytokine secretion. The results demonstrated that UMRA is a homogeneous acidic polysaccharide with a weight-average molecular weight of approximately 37.66 kDa. It comprised with rhamnose, galacturonic acid, galactose, glucuronic acid, arabinose, and mannose in a molar ratio of 3.94∶3.34∶2.26∶1.00∶0.95∶0.46. Further structural analysis revealed that UMRA possesses a pectic backbone with an rhamnogalacturonan-I  framework, featuring a repeating [→2)-α-L-Rhap-(1→4)-α-D-GalpA-(1→] disaccharide unit as its main chain, with side chains consisting of galactose, arabinose and glucuronic acid. UMRA exhibited no cytotoxicity toward macrophages at concentrations ranging from 0.4 to 250 μg/mL and promoted macrophage secretion of nitric oxide (NO), tumor necrosis factor-α (TNF-α), and interleukin-6 (IL-6). These findings provide a foundation for the structural elucidation and potential pharmaceutical development of polysaccharides derived from U. macrorrhiza.
