藏花素对H2O2诱导PC12细胞Bcl-2、Bax和Caspase-3的影响

Effects of Crocin on Bcl-2, Bax and Caspase-3 Expression of PC12 Cells Injured by H2O2

摘要: 本文通过观察藏花素对H2O2诱导的PC12细胞的保护作用和对Bcl-2、Bax和Caspase-3的影响,探讨藏花素对阿尔茨海默病细胞模型保护作用的机制。MTT法及LDH活性测定观察藏花素对PC12细胞模型的影响,RT-PCR检测Bcl-2、Bax和Caspase-3 mRNA表达水平,Western blot检测Bcl-2、Bax和Caspase-3的蛋白表达。藏花素预保护浓度在0.625 μM和5 μM之间,细胞存活率随着藏花素浓度的升高而升高;藏花素组和VE组与模型组相比,Bcl-2 mRNA和蛋白表达增加(P<0.05),Bax和Caspase-3 mRNA和蛋白表达降低(P<0.05)。研究结果表明:藏花素可能是通过上调Bcl-2的表达,从而介导PC12细胞发挥抗氧化和细胞凋亡的生物学功能。

Abstract: In this study,the effects of crocin on Bcl-2, Bax and Caspase-3 expression of PC12 cells injured by H2O2 were investigated.The possible mechanism of its protective effect on Alzheimer’s disease (AD) model cells was further explored.The effect of crocin on PC12 cells was detected using MTT and LDH assays.The expression of mRNA and protein (Bcl-2, Bax and Caspase-3) were detected by RT-PCR and Western Blot assays.With the concentration of crocin at 0.625 μM and 5 μM,the cell survival rate increasing in a dose-dependent manner.The expression levels of mRNA and protein of Bcl-2 increased,while expressions of Bax and Caspase-3 were decreased in crocin group and VE group (compared with the model group, P<0.05).These results indicated that the antioxidant and antiapoptosis effects of crocin were possible induced through increasing expression of Bcl-2 in PC12 cells.
