大高良姜地下根茎挥发油化学成分及体外药理活性研究

摘要:

采用水蒸气蒸馏法提取大高良姜地下根茎挥发油,通过气相色谱-质谱联用(GC-MS)分析其化学成分,分别采用DPPH、ABTS、抑菌圈、肉汤稀释及MTT等方法评价其体外药理活性。结果表明,挥发油主要成分为1,8-桉叶油素(25.41%)、β-没药烯(6.24%)和β-倍半水芹烯(9.08%)。挥发油对ABTS自由基有一定的清除活性,其抗坏血酸当量为4.83 mg AEs/g oil,对DPPH自由基显示出较差的清除活性。挥发油对金黄色葡萄球菌、枯草芽孢杆菌和变形杆菌表现出较好的抗菌活性,其抑菌圈大小为9.81~14.95 mm,MIC值为312.5~625 μg/mL,MBC值为625~1 250 μg/mL。挥发油具有较强的抗肿瘤活性,其对A549、K562和PC-3肿瘤细胞的IC50值分别为:26.57、58.01和35.56 μg/mL,与正常细胞相比具有显著性差异(P<0.05)。挥发油对酪氨酸酶、α-葡萄糖苷酶、乙酰胆碱酯酶和丁酰胆碱酯酶表现出一定的酶抑制活性,其阳性当量值分别为71.88±2.33 mg AREs/g oil、0.25±0.002 mmoL ACEs/g oil、0.77±0.02 mg GALAEs/g oil和0.52±0.01 mg GALAEs/g oil。因此,大高良姜地下根茎挥发油具有一定抗氧化活性、抗菌活性、酶抑制活性和较强的抗肿瘤活性。研究结果可为大高良姜地下根茎挥发油的进一步开发利用提供一定的数据参考。

Abstract:

The essential oil from the rhizome of Alpinia galanga (L.) Willd.was extracted by hydrodistillation,and the chemical components were analyzed by GC-MS.The in vitro pharmacological activities of essential oil were evaluated by DPPH,ABTS,the inhibition zone,the broth microdilution and MTT methods,and so on.The results showed that the predominant components of the essential oil were 1,8-cineole (25.41%),β-bisabolene (6.24%),and β-sesquiphellandrene (9.08%).The essential oil revealed a certain ABTS radical-scavenging activity with the equivalents of ascorbic acid was 4.83 mg ACEs/g oil,and revealed a weak DPPH radical-scavenging activity.The essential oil exhibited a significant antimicrobial activity against Staphylococcus aureus,Bacillus subtilis,and Proteus vulgaris with the inhibition zones (9.81-14.95 mm),MIC (312.5-625 μg/mL),and MBC (625-1 250 μg/mL).The essential oil showed significant cytotoxicity against human leukemic (K562),lung cancer (A549),and prostatic carcinoma (PC-3) cell lines with the IC50 values of 26.57,58.01 and 35.56 μg/mL,respectively.Compared with normal cells,there was a significant difference (P<0.05).The essential oil showed a certain enzyme inhibitory activity against tyrosinase,α-glucosidase,acetylcholinesterase,and butyrylcholinesterase with the equivalents of positive drugs were 71.88±2.33 mg AREs/g oil,0.25±0.002 mmoL ACEs/g oil,0.77±0.02 mg GALAEs/g oil,and 0.52±0.01 mg GALAEs/g oil,respectively.Therefore,the essential oil from rhizome of A. galanga has certain antioxidant activity,antibacterial activity,enzyme inhibition activity,and strong antitumor activity.The results can provide some data references for the further development and utilization of the essential oil from the rhizome of A. galanga.
