长叶紫珠化学成分及抗炎活性研究

Chemical constituents from Callicarpa longifolia and their anti-inflammatory activity

摘要:

本文对长叶紫珠(Callicarpa longifolia)进行化学成分研究。运用硅胶、Sephadex LH-20、ODS、MCI等柱层析并结合半制备型HPLC、重结晶等手段对长叶紫珠的甲醇提取物进行分离纯化;通过显色反应、理化性质、核磁共振波谱技术、质谱法等对所分离得到的化合物进行结构鉴定,从长叶紫珠中分离得到10个化合物,分别为18-hydroxyferruginol(1)、dehydroabietanol(2)、neolupenol(3)、kaempferol-3,4′,7-O-trimethylether(4)、artemein(5)、5-羟基-7,8,3′,4′-四甲氧基黄酮(6)、penduleti(7)、毛蕊花糖苷(8)、邻苯二甲酸二(2-乙基己基)酯(9)、邻苯二甲酸二丁酯(10)。十个化合物均为首次从长叶紫珠植物中分离得到。运用Griess法测定化合物1~58~10对LPS诱导的RAW 264.7细胞模型NO释放量的影响研究其抗炎活性。结果显示,化合物12、4、10具有一定的抑制NO生成作用,其IC50值分别为25.2、34.6、109.6、104.8 μmol/L。

Abstract:

To study the chemical constituents of Callicarpa longifolia.The methanol extract of C. longifolia was isolated and purified by silicon,Sephadex LH-20,ODS,MCI column chromatographies,semi-preparative HPLC and recrystallization method.The structures of the compounds were identified by color reaction,physicochemical properties,nuclear magnetic resonance spectroscopy and mass spectrometry.Ten compounds,18-hydroxyferruginol (1),dehydroabietanol (2),neolupenol (3),kaempferol-3,4′,7-O-trimethylether (4),artemiin (5),5-hydroxy-7,8,3′,4′-tetramethoxyflavone (6),penduleti (7),verbascoside (8),di-(2-ethylhexyl)-phthalate (9),and dibutyl phthalate (10) were isolated from C. longifolia for the first time.The anti-inflammatory effect of compounds 1-5 and 8-10 was tested on lipopolysaccharide-stimulated RAW 264.7 macrophages.The results showed that compounds 1,2,4 and 10 had significant inhibitory effects on NO production and their IC50 values were 25.2,34.6,109.6 and 104.8 μmol/L respectively.
