基于UPLC-Q-TOF-MS/MS的裸花紫珠水提物化学成分分析

Study on chemical components of aqueous extract from Callicarpa nudiflora by UPLC-Q-TOF-MS/MS

摘要: 为了对裸花紫珠水提物的化学成分进行分析鉴定,本研究以加热回流提取法制备裸花紫珠水提物,并采用超高效液相色谱串联四级杆飞行时间质谱法(UPLC-Q-TOF-MS/MS)通过高分辨质谱数据并结合相关文献,分析鉴定水提物的化学成分。最终从裸花紫珠药材水提物中共鉴定出37个化学成分,包括5个黄酮类化合物,16个苯乙醇苷类化合物及16个环烯醚萜类化合物。其中,京尼平苷酸、连翘脂苷E、肉苁蓉苷F为首次从裸花紫珠中报道。并推测木犀草苷、连翘酯苷B、毛蕊花糖苷和异毛蕊花糖苷为裸花紫珠的标志性成分,可为裸花紫珠后续药效物质基础研究及中成药二次开发和质量控制提供参考。

Abstract:

In order to analyze and identify the chemical composition of Callicarpa nudiflora aqueous extract,the aqueous extracts were prepared by heating reflux extraction and analyzed and identified by ultra-performance liquid chromatography-quadrupole-time of flight-mass spectrometry (UPLC-Q-TOF-MS/MS) through high-resolution mass spectrometry data and relevant literature.Finally,a total of 37 chemical constituents were identified from the aqueous extract of C. nudiflora,including 5 flavonoids,16 phenylethanoid glycosides and 16 iridoids.Among them,three compounds were reported for the first time from C. nudiflora,named geniposidic acid,forsythoside E and cistanoside F.Simultaneously,it was hypothesized that luteoloside,forsythiaside B,verbascoside and isoverbasin glycosides are the potential Q-markers,which can provide references for the subsequent study of medicinal material basis,secondary development,and quality control of C. nudiflora.
