芸香化学成分及其促黑色素生成作用研究

摘要:

为研究芸香(Ruta graveolens L.)化学成分其促进黑色素生成活性,采用硅胶、Sephadex LH-20 等多种色谱分离技术从芸香地上部分乙醇提取物乙酸乙酯萃取层中分离得到23个化合物,通过质谱和核磁共振等波谱技术对其结构进行鉴定,分别为布卢门醇C葡糖苷(1)、玫瑰花糖苷(2)、8-hydroxysmyrindiol(3)、cidioside B(4)、繸状芸香苷酯(5)、异芸香呋喃香豆醇葡萄糖苷(6)、芸香扔(7)、没食子酸(8)、对羟基苯甲酸(9、cis-p-coumaric acid 4-O-β-D-glucopyranoside(10)、2-苯乙基 6-O-没食子酸基-β-D-葡萄糖苷(11)、原儿茶酸(12)、6-羧乙基-5-羟基-苯并呋喃-5-O-β-D-吡喃葡萄糖苷(13)、picraquassioside A(14)、桑色素-3-O-α-L-鼠李糖苷(15)、山奈酚-3-O-β-D-6-乙酰基-吡喃葡萄糖-(1→4)-α-L-吡喃鼠李糖苷(16)、山奈酚(17)、槲皮素-3-O-α-L-吡喃鼠李糖苷(18)、槲皮素(19)、槲皮素-3-O-β-D-木糖苷(20)、山柰酚-3-O-α-L-吡喃鼠李糖苷(21)、槲皮素-3-O-(6″-O-反式香豆酰基)-β-D-葡萄糖苷(22)、柚皮素-7-O-β-D-葡萄糖苷(23)。其中1~51011131516181921~23首次从芸香中分离得到。化合物1~7处理小鼠B16黑色素瘤细胞,以细胞内黑色素含量,酪氨酸酶活性为评价指标,筛选其促进黑色素生成活性,经过结果分析:与溶剂对照组相比,化合物处理组并未表现出显著促进黑色素生成活性。

Abstract:

The chemical constituents of the aerial part of Ruta graveolens L.and their B16 melanin promotion activity have been studied.The isolation was carried out using chromatographic analyses including silica gel,Sephadex LH-20,etc.As a result,23 compounds were isolated.Their structures were elucidated as blumentol C glucoside (1),(6R,9S)-roseoside (2),8-hydroxysmyrindiol (3),cnidioside B (4),rutarensin (5),isorutarin (6),rutarin (7),gallic acid (8),p-hydroxybenzoic acid (9),cis-p-coumaric acid 4-O-β-D-glucopyranoside (10),2-phenylethyl 6-O-galloyl-β-D-glucopyranoside (11),protocatechuic acid (12),6-carboxyethyl-5-hydroxy-benzofuran 5-O-β-D-glucopyranoside (13),picraquassioside A (14),morin-3-O-α-L-rhamnopyranoside (15),kaempferol-3-O-β-D-6-acetyl glucopyranosyl-(1→4)-α-L-rhamnoside (16),kaempferol (17),quercitrin-3-O-α-L-rhamnoside (18),quercetin (19),quercetin-3-O-β-D-xylopyranoside (20),kaempferol-3-O-α-L-rhamopyranoside (21),helichrysoside (22),naringenin-7-O-β-D-glucopyranoside (23) on the basis of spectroscopic analysis.Compounds 1-5,10,11,13,15,16,18,19 and 21-23 were isolated from this plant for the first time.Compounds 1-7 of melanin contents and tyrosinase activity were performed in B16 melanoma cells.The results showed that,all compounds did not show significant melanin promotion activity compared with control group.
