Correlation between Cytotoxic Activity and HPLC Fingerprint Chromatogram of the Effective Fractions of Radix Pseudostellariae

Abstract: MTT assay was applied to screen the cytotoxic activities of the fractions of Radix Pseudostellariae (RP) on cell lines of MGC80-3, HepG2 and RKO. Cyclopeptides of RP were identified by TLC. And the correlation between cytotoxic activity and the HPLC fingerprints of the effective fractions was analyzed. The results showed that the ethyl acetate fraction had inhibitory effect on MGC80-3 and HepG2 cell lines. Furthermore, the fraction G which isolated from the ethyl acetate fraction exhibited significant cytotoxicity against all these three tumor cell lines. Comparison the HPLC and TLC fingerprints of the ethyl acetate fraction with those of fraction G suggested that the coefficient of all characteristic constituents resulted in their cytotoxic effect. In addition, the effect may be correlated with the existence of cyclopeptides.
