Abstract: Isolation and purification of antifungal substance from Paenibacillus polymyxa HY96-2, a commercial biocontrol strain used in control of fusarium wilt disease was studied in this paper. By means of acidification, n-BuOH extraction, ethyl acetate precipitation, silica gel column chromatography, sephadex LH-20 column chromatography and HPLC preparation, 6B was isolated and purified. Its structure was elucidated as fusaricidin A by means of spectroscopic analysis including NMR, MS and ESI-MS/MS. Results of two-fold serial agar dilution method showed that the minimum inhibition concentrations of 6B against 15 phytopathogenic fungi 12.5~50 µg/mL. Greenhouse results showed that 6B had better control effect on Botrytis cinerea of cucumber. The control effect could reach 95% when its concentration was set at 250 μg/mL.
