Preparative Separation of Two glycosides from Swertia Mussotti by High-Speed Counter-Current Chromatography

Abstract: A high performance method for isolation and purification of two glycosides from Swertia mussotti was successfully established by utilizing high-speed counter-current chromatography (HSCCC). Separation was performed with a two-phase solvent system composed of n-butanol-chloroform-methanol-water (3.4:8:5:6, v/v). 1-O-primeverosyl-3,7,8-trimethoxyxanthone (11 mg) and isoorientin (24 mg) were isolated and purified in one-step elution from 100 mg crude sample under the conditions of a flow rate 1.5 mL/min in the initial 210 min followed by 2.5 mL/min, 800 r/min and the detection wavelength of 254 nm. HPLC analysis showed that the purity of two compounds was over 99%. Their chemical structures were confirmed by UV, 1H- and 13C-NMR.
