不同品种芹菜叶中精油的提取及其功效测定

Extraction and Bioactivity of Essential Oil from Apium graveolens L.Leaves

摘要: 以杭州本地旱芹的两种品种的芹菜叶为原料提取芹菜叶精油,并设计正交试验确定提取芹菜叶精油的最佳条件,测定精油的抗菌作用及抗氧化作用。采用水蒸气蒸馏法提取芹菜叶精油,最佳提取条件采用正交试验的方法进行考察;采用平板法及抑菌圈法测定精油的抗菌作用;采用DPPH清除自由基法、铁离子还原法测定精油的抗氧化作用。实验结果显示最佳提取条件为:料液比1:8 g/mL,浸泡时间0.5 h,蒸馏时间3 h,在最佳提取条件下,两种芹菜叶精油有着相近的提取率;体外抑菌试验表明,芹菜叶精油对金黄色葡萄球菌和枯草芽孢杆菌具有明显的抑制作用,而对大肠杆菌无明显的抑菌作用;芹菜叶精油具有较低的抗氧化作用,其作用低于抗坏血酸和二丁基羟基甲苯(BHT)。

Abstract: Apium graveolens L.leaves essential oil was extracted from two kinds of Hangzhou local celeries.To test the antibacterial activity and antioxidant effect of A.graveolens leaves,the orthogonal experiment was performed to determine the optimal conditions for the extraction of essential oil from A.graveolens leaves.The spread plate method and bacteriostatic circle method were introduced to measure the antimicrobial activity of A.graveolens leaves essential oil.DPPH free radical assay and Fe3+-reducing assay were employed to measure the antioxidant activity of A.graveolens leaves essential oil.The results showed that the optimal extraction conditions were as follows:with solid/liquid ratio of 1:8 g/mL,0.5 h of soaking and 3 h of distillation. A.graveolens leaves essential oil had significant inhibitory effects on Staphylococcus aureus and Bacillus subtilis, but no effects on Escherichia coli. Compared with Vc and butylated hydroxytoluene (BHT),A.graveolens leaves essential oil had lower antioxidant effects.
