苦荞麦总黄酮对PA诱导脐静脉内皮细胞DDAH2表达的影响

摘要: 观察苦荞麦总黄酮对软脂酸(PA)诱导的人脐静脉内皮细胞(EA.hy926)中二甲基精氨酸二甲胺水解酶Ⅱ(DDAH2)表达等指标的影响,探讨苦荞麦总黄酮在胰岛素抵抗信号传导通路中的作用机制。将EA.hy926细胞株常规培养、传代,部分用600 μmol/L PA造模后分为对照组、模型组、苦荞麦总黄酮组和二甲双胍组,苦荞麦总黄酮组进一步加入浓度为125 mg/L的苦荞麦总黄酮,二甲双胍组加入浓度为2 mmol/L的二甲双胍。双抗体夹心法测定ADMA含量;硝酸还原酶法测定NO含量;RT-PCR法测定DDAH2 mRNA表达;Western blot测定DDAH2蛋白表达。苦荞麦中、高剂量组与模型组比较,ADMA含量明显减少,NO含量明显增加,均有显著性差异(P<0.05);苦荞麦总黄酮组与模型组比较,DDAH2 mRNA和蛋白表达量增加,有显著性差异(P<0.05)。苦荞麦总黄酮对于PA诱导下 EA.hy926细胞中DDAH2、NO的合成具有明显促进作用,对ADMA的合成有明显抑制作用。

Abstract: To observe the effects of total flavonoids of Fagopyrum tataricum (L.) Gaertn (tartary buckwheat) on DDAH2 in EA.hy926 cells in the presence of palmitic acid (PA),EA.hy926 cells were divided into control group,insulin resistance group,tartary buckwheat total flavonoids group and metformin group.Each group were combined insulin (50 nmol/L).Compared with control group,other groups were combined palmitic acid (600 μmol/L).Tartary buckwheat total flavonoids group was combined total flavonoids of tartary buckwheat (125 mg/L).Metformin group was combined metformin (2 mmol/L).The contents of ADMA were tested by double antibody sandwich method.The contents of NO were tested by nitrate reductase method.The DDAH2 mRNA and protein expression levels were determined by RT-PCR and Western-blot assays,respectively.Compared with insulin resistance group,the content of ADMA of middle and high dose groups were significantly reduced,the content of NO were increased significantly (P<0.05).Compared with insulin resistance group,the expression levels of DDAH2mRNA and protein were significantly higher in tartary buckwheat group (P<0.05).Total flavonoids of tartary buckwheat promoted the expression of DDAH2 mRNA and protein effectively in EA.hy926 cells under PA stimulation
