巴卡亭III转化生成10-脱乙酰基巴卡亭III的微生物筛选与鉴定

Screening and Identification of Strains Transforming Baccatin III into 10-Deacetyl Baccatin III

摘要: 10-脱乙酰基巴卡亭III(10-DAB)是紫杉烷类抗肿瘤药物多烯紫杉醇的前体物质。以巴卡亭III为底物,通过TLC、HPLC、LC-MS等方法,对课题组前期从污泥中得到的分离株进行筛选,获得了六株转化巴卡亭III生成10-DAB的菌株,分别命名为P1-A-19、P2-A-8、P2-A-10、P2-A-11、P2-A-12、P2-A-13,通过形态学观察、生理生化试验和16S rDNA序列分析确定其种属信息,P1-A-19被鉴定为解鸟氨酸拉乌尔菌(Raoultella ornithinolytica),P2-A-8被鉴定为成团泛菌(Pantoea agglomerans),P2-A-11、P2-A-12、P2-A-13均被鉴定为铜绿假单胞菌(Pseudomonas aeruginosa)。其中,Raoultella ornithinolytica P1-A-19是首次发现的产10-DAB能力的微生物。

Abstract: 10-Deacetyl baccatin III(10-DAB)is the synthetic precursor of the taxanes drug docetaxel with anti-tumor activity.By the biotransformation of baccatin III,six 10-DAB producing strains isolated from the sludge were screened by the combinational methods of TLC,HPLC and LC-MS,which were named as P1-A-19,P2-A-8,P2-A-10,P2-A-11,P2-A-12 and P2-A-13,respectively.Combining with morphological observation,physiological and biochemical characteristics,16S rDNA sequence analysis,P1-A-19 was identified as Raoultella ornithinolytica, P2-A-8 was identified as Pantoea agglomerans, P2-A-10,P2-A-11,P2-A-12,P2-A-13 were identified as Pseudomonas aeruginosa. Raoultella ornithinolytica  P1-A-19 was firstly detected to produce 10-DAB from baccatin III.
