基于UPLC-MS/MS法研究淫羊藿属植物品种品质与生长海拔、植物类群间关系

摘要: 应用UPLC-MS/MS法对10种淫羊藿属植物主成分含量差异进行比较研究,以探讨药材品种品质与生长海拔、植物类群间联系。采用Phenomenex C18柱(50 mm×2.1 mm,3 μm),以乙腈-0.1%甲酸水为流动相梯度洗脱,流速为0.2 mL/min;质谱离子源为ESI,多反应监测正离子模式进行检测。淫羊藿苷、朝藿定A、朝藿定B、朝藿定C分别在0.995~9950.000、0.992~9920.000、1.021~10210.000、1.035~10350.000 ng/mL浓度范围内显示出良好的线性关系(r≥0.9996),平均加样回收率为98.7%~101.2%,RSD为1.7%~2.4%;结果表明,不同品种淫羊藿主成分含量差异较大,且含量有随海拔升高而降低的趋势,小花类群各成分含量均明显高于大花。本研究证明了品种品质与生长海拔、植物类群间具有相关性;该方法准确、快速、灵敏度高,适用于淫羊藿药材的质量控制。

Abstract: In this study,the content of the main components in Epimedium was compared by UPLC-MS/MS analysis to explore the correlation between Epimedium quality, growth altitude and plant groups.The analysis was performed on a Phenomenex-C18 column (50 mm×2.1 mm,3 μm).The mobile phase consisted of acetonitrile (B) and 0.1% formic acid (A) were used for gradient elution.The flow rate was 0.2 mL/min.The ion source of MS were ESI with multiple reaction monitoring mode.A good linear relationship between icariin,epidemin A,epidemin B and epidemin C were found in the concentration range of 0.9959950.000,0.992-9920.000,1.021-10210.000 and 1.035-10350.000 ng/mL,respectively.The average recovery rate were 98.7%-101.2% and the RSD were 1.7%-2.4%.The result showed that the content of main components were significantly different in different species of Epimedium, and the content decreased with the increasing of elevation.The content of each components in small flowered taxa of Epimedium were significantly higher than the small flowered taxa.This methods were accurate,rapid,highly sensitive and suitable for the qualitative analysis of main component of Epimedium.
