山楂总黄酮对心肌缺血大鼠尿液代谢谱的影响

Effect of Hawthorn Flavonoids on 1H NMR Urine Metabolism of Myocardial Ischemia Rats

摘要: 为了研究山楂总黄酮对异丙肾上腺素(ISO)诱导的心肌缺血大鼠尿液代谢谱的影响及其代谢通路,取雄性SD大鼠24只,随机分为正常组、模型组和山楂总黄酮组(0.276 g/kg),灌胃给药,连续5天。1H NMR检测大鼠尿液代谢物变化,并进行模式识别分析。确认了6个生物标志物,山楂总黄酮(0.276 g/kg)上调TMAO、Creatinine(P < 0.05),下调DMA、DMG、Hippurate、Citrate(P < 0.05)。通路分析表明山楂总黄酮可能通过调节钙超载、氧化应激、三羧酸循环以及肾功能改善心肌缺血。本文为在细胞和分子水平阐释山楂总黄酮抗心肌缺血的作用机制提供了参考,为筛选山楂总黄酮中的抗心肌缺血成分提供了代谢组学分析手段。

Abstract: The aim of the present study was to investigate the effect of Hawthorn flavonoidson for myocardial ischemia rats induced by isoproterenol,and subsequently discuss the different metabolic pathway.1H NMR was used to exam the urine metabolites of rats,the pattern identification were adapted to revealed potential biomarkers in urine.There were significant differences in urine metabolies among the groups,six urinary metabolic products were compareded with the control group.After the treatment of hawthorn flavonoids (0.276 g/kg),the levels of DMA,DMG,Hippurate and citrate decreased (P < 0.05),TMAO and Creatinine increased (P < 0.05).Myocardial Ischemia Rats showed significant difference metabolic profile compared with the normal group.Hawthorn flavonoids (0.276 g/kg) changed the myocardial Ischemia metabolic profile to the normal status in some extent possibly through its regulation on the oxidative stress,calcium overload of cells,tricarboxylic acid cycle as well as the renal function.
