摘要: 采用超高效液相色谱-串联四级杆-飞行时间质谱(UPLC-QTOF-MS)技术快速识别和鉴定了野生与栽培杨树桑黄子实体的化学成分。选用反相Waters Acouity UPLC BEH C18(100 mm×2.1 mm,1.7 μm)色谱柱,甲醇与水(含0.1%甲酸)作为流动相二元线性梯度洗脱,在电喷雾负离子模式下扫描化合物的分子离子峰、二级质谱碎片信息,并结合保留时间和相关文献,从野生和栽培杨树桑黄子实体中共鉴定出40个化合物,包括小分子有机酸8个、儿茶素类3个、吡喃酮类20个、黄酮9个,其中野生和栽培子实体中共有成分14个,野生子实体中特有成分14个,栽培子实体中特有成分12个。分析结果表明吡喃酮酚类化合物是杨树桑黄子实体中的主要活性成分之一,本研究可为野生和栽培杨树桑黄子实体的快速鉴定、质量控制以及阐明药效物质基础提供参考。

Abstract:

 The chemical compositions in wild and cultivated fruiting body of Sanghuangporus vaninii were analyzed rapidly by ultra performance liquid chromatography quadrupole time-of-flight mass spectrometry (UPLC-QTOF-MS).Waters Acouity UPLC BEH C18 (100 mm×2.1 mm,1.7 μm) chromatographic column and methanol with water (containing 0.1% formic acid) gradient elution were used to analyze the extracts sample.The ESI-MS negative molecular ion peaks,MS2 product ions,retention time and together with the references were used to tentative identify the chemical constituents.The results indicated that 40 metabolites from wild and cultivated fruiting bodies of S. vaninii were identified,including 8 organic acids,3 catechins,20 pyranones and 9 flavonoids,of which 14 components in wild and cultivated fruiting body,14 unique components in wild and 12 unique components in cultivated fruiting body.The analysis results showed that pyranone phenolic compounds were presumed to be mainly active components in the fruiting body of S. vaninii. This method is rapid,sensitive,which can provide the reference for evaluation of the quality from S. vaninii.
