长管红山茶叶片的化学成分研究

摘要:

为研究长管红山茶(Camellia longituba Chang)叶片的化学成分,采用正相硅胶、反相硅胶、凝胶色谱、半制备高效液相色谱仪(HPLC)和高速逆流色谱仪(HSCCC)等一系列方法进行分离纯化,得到15个化合物,根据理化性质结合波谱数据分别鉴定为对羟基肉桂酸(1)、槲皮素-3-O-β-D-半乳糖苷(2)、槲皮素(3)、(+)-儿茶素(4)、原儿茶酸(5)、5,7-二羟基色原酮(6)、芦丁(7)、槲皮素-3-甲醚(8)、9-羟基-4-甲氧基补骨脂素(9)、去甲丁香色原酮(10)、柚皮素(11)、山柰酚(12)、(-)-表儿茶素(13)、没食子酸(14)、杨梅素(15)。所有化合物均为首次从该植物中分离得到,且化合物1、67891011是首次从山茶属中分离得到。用红细胞裂解模型对化合物1~15进行尿素通道蛋白B(urea transporter B,UT-B)抑制活性筛选,结果显示化合物1415具有较强的抑制活性,其IC50值分别为2.373、2.521 μg/mL。

Abstract:

To study the chemical constituents from the leaves of Camellia longituba Chang.The chemical constituents from the leaves of C. longituba were isolated and purified by normal phase silica gel,reverse phase silica gel,sephedx LH-20 gel,semi-preparative HPLC and HSCCC.The compounds were identified by color,morphology,UV and NMR.Their structures were elucidated on the basis of physico-chemical properties and spectral data.15 compounds were isolated and identified as p-hydroxycinnamic acid (1),quercetin-3-O-β-D-galactoside (2),quercetin (3),catechin (4),protocatechuic acid (5),5,7-dihydroxychromone (6),rutin (7),3-O-methyl quercetin (8),xanthotoxin (9),noreugenin (10) ,naringin (11),kaempferol (12),epicatechin (13),gallic acid (14) and myricetin (15).All compounds were isolated from C. longituba for the first time,and compounds 1,6,7,8,9,10 and 11 were isolated from Camellia for the first time.Compounds 1-15 were screened for urea transporter B (UT-B) inhibitors using red blood cell lysing model.The results showed that compounds 14 and 15 have strong inhibitory activities,with IC50 values of 2.373 and 2.521 μg/mL,respectively.
