马鞭草药材UPLC指纹图谱建立及指标性成分的测定

摘要:

建立马鞭草药材指纹图谱,并对4种有效成分的含量进行测定,为马鞭草药材质量控制提供参考。采用超高效液相色谱(UPLC)法建立马鞭草药材指纹图谱,并利用高分辨质谱对共有峰进行指认,以15批马鞭草药材指纹图谱共有峰的相对峰面积为变量,进行化学计量学分析,并对15批马鞭草药材有效成分戟叶马鞭草苷、马鞭草苷、毛蕊花糖苷和异毛蕊花糖苷的含量进行测定。结果显示,马鞭草药材指纹图谱有16个共有峰,并指认出其中6个成分,聚类分析(HCA)和主成分分析(PCA)将15批药材分为3类,正交偏最小二乘法-判别式分析(OPLS-DA)共筛选出7个VIP>1.0的差异性成分。含量测定结果显示,不同产区的马鞭草药材戟叶马鞭草苷、马鞭草苷、毛蕊花糖苷和异毛蕊花糖苷的含量存在较大的差异。该方法能有效分析不同产地马鞭草药材的质量差异,为不同产地马鞭草药材质量的评价提供参考。

Abstract:

The fingerprint of Verbenae Herba was established and the contents of four effective components were determined to provide reference for the quality control of Verbenae Herba.The fingerprint of Verbenae Herba was established by ultra performance liquid chromatography (UPLC )method,and the common peaks were identified by high resolution mass spectrometry.The relative peak areas of the common peaks in the fingerprints of 15 batches of Verbenae Herba were used as variables for chemometric analysis,and the contents of the effective components of hastatoside,verbenoside,acteoside and isoacteoside in 15 batches of Verbenae Herba were determined.The results showed that there were 16 common peaks in the fingerprint of Verbenae Herba,and six of them were identified.Cluster analysis (HCA) and principal component analysis (PCA) divided 15 batches of medicinal materials into three categories,and orthogonal partial least squares discriminant analysis (OPLS-DA) screened seven different components with VIP>1.0.The content determination results showed that the contents of hastatoside,verbenoside,acteoside and isoacteoside in different production areas were significantly different.This method can effectively analyze the quality difference of Verbenae Herba from different habitats,and provide a reference for the quality evaluation of Verbenae Herba from different habitats.
