黄芪汤多成分含量测定及其抗非酒精性脂肪肝病作用的谱效关系研究

Determination of multiple components in Huangqi decoction and its spectrum-effect relationship against non-alcoholic liver disease

摘要:

运用高效液相色谱-二极管阵列检测器-蒸发光散射检测器(high-performance liquid chromatography coupled with diode array and evaporative light scattering detectors,HPLC-DAD-ELSD)联用技术对黄芪汤多成分进行含量测定,探讨黄芪汤对非酒精性脂肪肝病的治疗作用及药效物质。利用HPLC-DAD-ELSD联用技术测定8个成分的含量,采用高脂饮食建立大鼠非酒精性肝损伤模型进行药效评价,并通过油酸诱导HepG2细胞建立体外脂肪堆积模型,通过双变量相关分析和灰色度关联分析对其降脂活性进行谱效分析,筛选黄芪汤治疗非酒精性肝损伤的药效物质。结果表明,8个成分毛蕊异黄酮葡萄糖苷、甘草苷、芒柄花苷、甘草素、毛蕊异黄酮、芒柄花素、甘草酸和黄芪甲苷质量分数分别为0.711±0.316、0.403±0.451、0.385±0.181、0.053±0.071、0.037±0.015、0.037±0.004、13.37±3.544和0.110±0.051 mg/g。体外谱效关系发现黄芪甲苷、甘草酸、芒柄花素、毛蕊异黄酮葡萄糖苷、芒柄花苷、芹糖甘草苷、甘草素、毛蕊异黄酮和甘草苷9个成分与黄芪汤降脂活性高度相关。该研究建立的黄芪汤治疗非酒精性脂肪肝多成分含量测定方法可准确、高效,同时黄芪汤有良好的防治非酒精性脂肪肝的疗效,可为黄芪汤保健品的开发提供参考。

Abstract:

This study aims to establish the content of multiple components in Huangqi decoction by High-performance liquid chromatography coupled with diode array and evaporative light scattering detectors (HPLC-DAD-ELSD), and discuss the therapeutic effects of Huangqi decoction on non-alcoholic liver disease. The contents of 8 active evaluation. Next, oleic acid-induced HepG2 were used as the model of lipid accumulation in vitro. The composition-activity relationship between active ingredients and the ingredients were determined by HPLC-DAD-ELSD. Non-alcoholic liver injury model was induced by a high-fat diet for efficacy cells lipid-lowering activity of Huangqi decoction was constructed by bivariate correlation analysis and gray relational analysis to screen pharmacodynamic substance. The content of the eight compounds were determined as follows: calycosin-7-O-β-D-glycoside (0.711±0.316 mg/g), liquiritin (0.403±0.451 mg/g), ononin (0.385±0.181 mg/g), liquiritigenin (0.053±0.071 mg/g), calycosin (0.037±0.015 mg/g), formononetin (0.037±0.004 mg/g), glycyrrhizic acid (13.37±3.544 mg/g), and astragaloside IV (0.110±0.051 mg/g). A total of nine compounds (astragaloside IV, glycyrrhizic acid, formononetin, calycosin-7-O-β-D-glycoside, ononin, liquiritin apioside, liquiritigenin, calycosin, and liquiritin) related to the lipid-lowering effect of Huangqi decoction. The established method for the determination of multi-component content of Huangqi Decoction in the treatment of non-alcoholic liver disease is accurate and efficient. Huangqi Decoction has a good effect on the treatment of non-alcoholic liver disease, which can be used as reference for the development of Huangqi decoction related health care products.
