姜黄素对棕榈酸诱导的C2C12细胞损伤的保护作用

Protective effects of curcumin on palmitic acid-induced injury in C2C12 cell

摘要:

探讨姜黄素(curcumin,Cur)对棕榈酸(palmitic acid,PA)诱导的C2C12细胞损伤的保护作用及其机制。通过建立PA诱导的损伤细胞模型,本研究检测了Cur对细胞活力和葡萄糖转运蛋白4(glucose transporter type 4,GLUT4)表达的影响,并采用油红O染色和甘油三酯测试试剂盒评估了Cur对脂代谢的影响。通过代谢组学分析探究了Cur对损伤细胞模型代谢谱的影响,并验证了其作用机制。结果显示,Cur能有效提高C2C12细胞活力,增加GLUT4表达水平,并降低细胞中脂质的积累。代谢组学和Western blot分析揭示Cur可能通过过氧化物酶体增殖物激活受体α(peroxisome proliferators-activated receptor α,PPARα) /酰基辅酶A氧化酶1(acyl-CoA oxidase 1,ACOX1)/肉碱棕榈酰转移酶1(carnitine palmitoyltransferase 1,CPT1)通路调节C2C12细胞的脂质代谢,从而发挥保护作用。研究结果为Cur在代谢性疾病治疗中的潜在应用提供了理论基础。

Abstract:

The aim of this study was to investigate the protective effect of curcumin (Cur) on palmitic acid (PA)-induced C2C12 cell injury and its mechanism.By establishing a cell model of PA-induced injury,this study examined the effects of Cur on cell viability and glucose transporter type 4 (GLUT4) expression,and assessed the effects of Cur on lipid metabolism using oil red O staining and a triglyceride test kit.The effect of Cur on the metabolic profile of the injured cell model was explored by metabolomics analysis,and its mechanism of action was verified.The results showed that Cur effectively increased C2C12 cell viability,increased GLUT4 expression levels,and decreased lipid accumulation in cells.Metabolomics and Western blot analysis revealed that Cur may regulate lipid metabolism in C2C12 cells through the peroxisome proliferators-activated receptor α (PPARα)/acyl-CoA oxidase 1 (ACOX1)/ carnitine palmitoyltransferase 1 (CPT1) pathway,thus exerting a protective effect.The results provide a theoretical basis for the potential application of curcumin in the treatment of metabolic diseases.
