星点设计—效应面法优化红梅消总皂苷双水相提取工艺

Optimization of Aqueous Two-Phase Extraction of Saponins from Rubus parvifolius L.by Modified Central Composite Design and Response Surface Methodology

摘要: 为得到红梅消中总皂苷的最佳提取工艺参数,首次采用硫酸铵-乙醇双水相体系对其进行提取;以双水相中两相的质量百分比为自变量,以红梅消总皂苷得率为因变量,通过确定中心点及缩小变量取值范围,用星点设计-效应面分析法进行优化,得到双水相最佳提取条件:硫酸铵12.14%,乙醇40.00%,总皂苷得率为2.95%。结果表明用星点设计-效应面分析法筛选双水相各相的质量百分比方法可行性高,结果可靠;双水相体系对红梅消总皂苷有较好的提取作用,工艺简单、节能、易放大于工业生产。

Abstract: The aqueous two-phase of ethanol/ammonium sulfate system was used for the extraction of saponins from Rubus parvifolius L..The extraction conditions were optimized using modified central composite design coupled with response surface methodology with two independent variables including quantity of (NH4)2SO4 and CH3COOH,and the saponin content as the dependent variable.The optimalconditions were determined as follows:12.14% (NH4)2SO4 and 40.00% CH3COOH. The extraction yield of saponins was 2.95%.The results showed that the central composite design and response surface methodology was useful for optimizing the aqueous two-phase extraction of saponins from R.parvifolius.The developed extraction technology was simple,energy-saving and easy to zoom in industrial production.
