制何首乌提取物及主要单体成分体外对酪氨酸酶活性的影响

Effect of the Extracts and Main Monomer Components of Processed Polygonum multiflorum on Activity of Tyrosinase in vitro

摘要: 采用蘑菇酪氨酸酶多巴速率氧化法,体外评价制何首乌水提取物、乙醇提取物各分离组分及大黄素,大黄素甲醚,大黄素-8-O-β-D-葡萄糖苷,2,3,5,4′-四羟基二苯乙烯-2-O-β-D-葡萄糖苷等制何首乌主要单体成分对体外酪氨酸酶活性的影响。结果未在制何首乌中发现能够体外有效激活酪氨酸酶活性的成分,制何首乌水提物,制何首乌乙醇提取物的50%乙醇洗脱物及95%乙醇洗脱物,2,3,5,4′-四羟基二苯乙烯-2-O-β-D-葡萄糖苷皆有浓度依赖的酪氨酸酶抑制活性,其中2,3,5,4′-四羟基二苯乙烯-2-O-β-D-葡萄糖苷在48 μg/mL的浓度下即可达到46.72%的抑制率,具有美白产品研发价值。

Abstract: This paper investigated the effect of water extract,separated parts of ethanol extract from processed Polygonum multiflorum and the monomer components including emodin, physcion, emodin-8-O-β-D-glucoside and 2,3,5,4′-tetrahydroxystilbene-2-O-β-D-glucoside (THSG) on the tyrosinase activity in vitro by measuring the oxidation rate of L-DOPA.The results showed that none of the tested substance could activate the tyrosinase activity effectively.Conversely,50% ethanol and 95% ethanol eluted components from the ethanol extract of processed Polygonum multiflorum, water extract of processed Polygonum multiflorum and THSG inhibited the tyrosinase activity in a concentration dependent manner.THSG exhibited a significant tyrosinase inhibitory activity (reached 46.72%) at the concentration of 48 μg/mL,which indicated that THSG has potential value in developing skin whitening product.
