甘松精油的盐析辅助水蒸气蒸馏法提取及其性质研究

Extraction of Nardostachys jatamansi DC. essential oil by salting-out assisted steam distillation and its property

摘要:

采用盐析辅助水蒸气蒸馏法(salting-out assisted steam distillation method,SSD)提取甘松精油,在单因素实验基础上,通过正交实验优化甘松精油提取的工艺条件。并考察氯化钠的加入对甘松精油得率、感官品质、物理性质(相对密度、酸值、酯值等)、化学成分、清除DPPH自由基能力的影响。采用SSD法提取甘松精油的最优提取条件是:粉碎度为50目,料液比为1∶12,提取时间为9 h,NaCl质量分数为5%。在此条件下,精油平均得率为4.08%,比水蒸气蒸馏法(steam distillation,SD)得率提高了8.2%。利用气相色谱-质谱联用技术对甘松精油的化学成分进行鉴定,共分离鉴定出56种化学成分,其主要成分是为白菖烯、桉叶-4(15),7-二烯-1β-醇、百秋李醇、(±)-杜松烯。加入氯化钠提取甘松精油,可有效提高精油提取率,对精油感官品质、化学成分、清除DPPH自由基能力影响较小,但对甘松精油的理化性质有一定影响。盐析辅助水蒸气蒸馏法提取甘松精油可提高甘松精油产率。

Abstract:

Salting-out assisted steam distillation method (SSD) was used to extract the essential oil of Nardostachys jatamansi DC..Based onsingle-factor experiments,the extraction conditions of essential oil from N. jatamansi.were optimized by orthogonal experiments.The effects of the addtion of sodium chloride on the essential oil′s yield,sensory quality,physical properties (relative density,acid value,ester value,etc.),chemical composition and scavenging ability of DPPH free radicals were investigated.The optimal extraction conditions for extracting the essential oil ofN. jatamansi by SSD method were:crushing degree 50 mesh,solid-liquid ratio 1∶12,extraction time 9 h,and NaCl mass fraction 5%.Under these conditions,the average yield of essential oil was 4.08 %,which was 8.2% higher than that of steam distillation (SD).The chemical components of the essential oil of N. jatamansi were identified by gas chromatography-mass spectrometry.A total of 56 chemical components were isolated and identified.The main components were calarene,eucalyptol-4 (15),7-diene-1β-ol,patchouli alcohol,(±)-cadinene.The addition of sodium chloride to extract N. jatamansi essential oil can effectively improve the extraction rate of essential oil and has little effect on the sensory quality,chemical composition and DPPH free radical scavenging ability of essential oil,but has a certain effect on the physical and chemical properties of N. jatamansi essential oil.Salting-out assisted steam distillation can improve the yield of essential oil of N. jatamansi.
