研究微波辅助提取法对西番莲(Passiflora edulis)果皮多糖的理化性质和功能活性的影响。分别制备微波辅助水提西番莲果皮多糖(microwave-assisted water extracted polysaccharide,MWP)、微波辅助酸提西番莲果皮多糖(microwave-assisted acid extracted polysaccharide,MAP)和微波辅助碱提西番莲果皮多糖(microwave-assisted lye extracted polysaccharide,MLP),并对三种提取方法提取的西番莲果皮多糖进行理化性质检测、结构表征和抗衰老活性研究。结果表明,MLP总糖含量最高(77.55%),MAP糖醛酸含量最高(71.11%),MAP的提取率最高(14.68%),MWP、MAP和MLP主要由半乳糖醛酸、甘露糖和葡萄糖组成,但占比有所不相同;多糖外观形貌、热稳定性和三螺旋结构方面均展现出差异。多糖均具有一定的抗氧化和抗衰老能力,MAP的活性最好,对DPPH自由基、羟基自由基和ABTS+自由基的清除率分别为87%、85%和66%,MAP浓度为400 μg/mL时,N2线虫最久可存活23 d,表现出较好的抗衰老活性。本研究可为西番莲果皮多糖在抗氧化和抗衰老方向的应用提供了理论依据,并促进西番莲果皮的资源再利用。

Abstract:

In order to investigate the impact of microwave-assisted extraction on the physical and chemical attributes and functional activities of Passiflora edulis peel polysaccharides, the microwave-assisted water extracted polysaccharides (MWP), microwave-assisted acid extracted polysaccharides (MAP), and microwave-assisted alkaline extracted polysaccharides (MLP) were prepared. The physical and chemical properties, structural characterization and anti-aging activity of the peel polysaccharides extracted using three different extraction methods were determined. The findings revealed that the MLP exhibited the highest total sugar content (77.55%), the MAP exhibited the highest uronic acid content (71.11%), and the MAP exhibited the highest extraction rate (14.68%). The predominant monosaccharides in the MWP, MAP, and MLP were galacturonic acid, mannose, and glucose, though the proportions differed. The polysaccharides exhibited diverse morphological characteristics, thermal stability and triple helix structures. All polysaccharides demonstrated antioxidant and anti-aging abilities, with MAP exhibiting the most pronounced activity. The DPPH free radical, hydroxyl free radical, and ABTS+ free radical scavenging rates of the MAP were 87%, 85%, and 66%, respectively. At a concentration of 400 μg/mL, the MAP exhibited anti-aging activity, enabling N2 nematodes to survive for 23 days and showed a good anti-aging activity. This study provides a theoretical basis for the application of peel polysaccharides of P. edulis in the direction of antioxidant and anti-aging, and promotes the reuse of P. edulis peel as a resource.
