Mechanism of Ziziphi Spinosae Semen oil in improving insomnia based on network pharmacology and experimental verification

摘要:

利用网络药理学和实验验证研究酸枣仁油(Ziziphi Spinosae Semen oil,ZSSO)改善失眠的作用机制。首先,利用气相色谱-质谱联用(gas chromatography-mass spectrometry,GC-MS)仪采集数据,结合文献和数据库鉴定ZSSO化学成分。然后,利用SwissTargetPrediction数据库获取ZSSO成分靶点,利用GeneCards、OMIM、DisGeNET数据库获取失眠疾病靶点。其次,利用Venny、Cytoscape 3.10.0、String和DAVID数据库分析ZSSO改善失眠的潜在活性成分、核心靶点及作用通路之间的关系。最后,通过建立大鼠失眠模型进行实验验证。ZSSO中共鉴定了27个化学成分。网络药理学结果显示ZSSO改善失眠的活性成分可能为油酸、亚油酸等4种化学成分;关键靶点可能为雄激素受体(androgen receptor,AR)、过氧化物酶体增殖物激活受体γ(peroxisome proliferator activated receptor gamma,PPARG)、γ-氨基丁酸A型受体β2亚基(gamma-aminobutyric acid type A receptor subunit beta2,GABRB2)、γ-氨基丁酸B型受体亚基1(gamma-aminobutyric acid type B receptor subunit1,GABBR1);这些靶点主要富集于神经活性配体-受体相互作用、环磷酸腺苷(cyclic adenosine monophosphate,cAMP)信号通路等多条信号通路。动物实验结果显示,ZSSO能明显增加大鼠血清和脑组织中5-羟色胺含量并增强脑内AR、PPARG蛋白表达。上述研究表明ZSSO具有改善失眠的作用,其作用机制可能与增强脑内AR、PPARG蛋白表达有关。

Abstract:

This study aims to investigate the mechanism of Ziziphi Spinosae Semen oil (ZSSO) in improving insomnia by using network pharmacology and experimental verification. Firstly, the gas chromatography-mass spectrometry (GC-MS) instrument was used to collect data, and the chemical components of ZSSO were identified from the existing literature and relevant databases. Next, the SwissTargetPrediction database was used to retrieve the targets of the ZSSO components. Meanwhile, the targets of insomnia were identified from the GeneCards, OMIM, and DisGeNET databases. Then the associations among potential active ingredients, key targets, and signaling pathways of ZSSO components were analyzed by the Venny, Cytoscape 3.10.0, String, and DAVID databases. And finally, the experimental results were validated by using a rat model of insomnia. The results of GC-MS analysis showed that twenty-seven chemical components were identified in ZSSO. The results of network pharmacology showed that the active ingredients of ZSSO in improving insomnia may be oleic acid, linoleic acid and other four ingredients, and the key targets may be androgen receptor (AR), peroxisome proliferator activated receptor gamma (PPARG), gamma-aminobutyric acid type A receptor subunit beta2 (GABRB2), and gamma-aminobutyric acid type B receptor subunit1 (GABBR1), which mainly enriched in neuroactive ligand-receptor interaction, cyclic adenosine monophosphate (cAMP) and other signaling pathways. The in vivo experimental results showed that the ZSSO could significantly increased the content of 5-hydroxytryptamine in serum and brain tissue, and enhanced the protein expression of AR and PPARG in the brain. The study suggested that the ZSSO can effectively improve insomnia, and the mechanism of action may be related to the enhancement of AR and PPARG in the brain.
